摘    要:目的:建立二维液相色谱/质谱法分析天然药物的技术平台。方法:以苦参生物碱为例,采用强酸性阳离子交换柱作预分离柱,柱后流动相修饰,使生物碱在富集柱上浓集,然后切换至分析柱系统,以反相柱,碱性流动相分离生物碱,用光二极管阵列检测器和质谱检测。从猪脑中提取的神经节苷脂,以氨基柱作预分离柱,反相色谱系统进行分离分析。结果:从苦参碱中检出了氧化苦参碱等5个生物碱;从神经节苷脂中检测出了单唾液酸神经节苷脂等5个成分。结论:已建立的分离分析系统高效、快速、自动化,可用于天然药物复杂体系的分离分析。

Abstract:AIM:To develop a 2D-LC/MS platform for analysis of natural products . METHOD:The matrine alkaloids were as one of the examples. The alkaloids were isolated and eluted by a strong cation exchange system. By using post column mobile phase modification techniques, the eluted alkaloids were trapped on top of a small C18 enrichment column, then the enrichment column was switched to the analytical column system to separate the alkaloids by using RP-column and basic mobile phase. DAD and mass spectrometer were used as detectors. Gangliosides were extracted from pig brain and were separated on a NH2 column in the first step and analyzed by a RP chromatographic system. RESULT:Five alkaloids were detected from matrine alkaloids and five compounds such as GMl were detected from the gangliosides. CONCLUTION: A highly efficient, high speed and automatic system has been established, which can be used to separate and analyze the complicated natural products.
