升陷汤治疗低血压性脑梗死的网络药理学作用机制研究

摘    要:目的探讨升陷汤治疗低血压性脑梗死的网络药理学作用机制。方法采用网络药理学的方法,在TCMSP数据库获取升陷汤有效成分,预测其靶点并收集;通过OMIM、GENECARD、DRUGBANK疾病相关靶点数据库检索低血压性脑梗死相关靶点信息。将药物靶点映射至疾病靶点集合上,获得交集基因,运用DAVID数据库对得到的交集进行GO和KEGG的富集分析,通过Cytoscape软件对结果进行可视化呈现。结果升陷汤主要通过干预低血压性脑梗死过程中炎症过程、凋亡途径及神经干细胞再生等过程,减轻低血压性脑梗死造成的神经损伤。其中,Akt1、TNF、RELA、IL-6、JUN、VEGFA、MYC、MAPK8、CXCL2、ESR1等为关键的调节位点。结论升陷汤治疗低血压性脑梗死主要通过调节炎症过程、凋亡途径及神经干细胞再生相关等途径。

Abstract:Objective:To investigate the network mechanism of Shengxian Decoction in the treatment of hypotensive cerebral infarction.Methods:Based on Network pharmacology,Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)was retrieved to obtain the active components and targets of Shengxian Decoction and to predict its targets and collect them.OMIM,GENECARD,and DRUGBANK databases were searched for the target information of hypotensive cerebral infarction.Drug targets were lined up with disease targets to obtain intersection genes.DAVID database was used to conduct GO and KEGG enrichment analysis of the resulting intersection genes.The results were visualized by Cytoscape software.Results:Shengxian Decoction alleviated the nerve damage caused by hypotensive cerebral infarction mainly through intervening the inflammatory process,apoptotic pathway,and neural stem cell regeneration.Akt1,TNF,RELA,IL-6,JUN,VEGFA,MYC,MAPK8,CXCL2,and ESR1 were the key targets.Conclusion:Shengxian Decoction can regulate the pathology of hypotensive cerebral infarction by affecting inflammatory process,apoptotic pathway,and neural stem cell regeneration.
