糖网明抗糖尿病大鼠视网膜神经组织Bax表达的研究

摘    要:目的:探讨凋亡相关基因Bax在早期糖尿病大鼠视网膜神经组织中的表达及糖网明的抗凋亡作用。方法:四氧嘧啶制作糖尿病大鼠模型,分为正常组、糖尿病未治疗组,糖尿病糖网明治疗组,180d后处死大鼠,制备视网膜切片免疫组化染色,并用原位末端标记法(TUNEL)法标记大鼠视网膜凋亡细胞,光镜观察。结果:糖尿病大鼠与正常组大鼠相比Bax基因的蛋白阳性反应增强,视网膜神经细胞凋亡数量显著升高,糖网明干预组与模型组相比这些改变明显较轻。结论:糖网明可抑制凋亡相关基因Bax的表达,抗视网膜神经细胞凋亡,对早期糖尿病视网膜病变具有良好的防治作用。

Abstract:Objective: To explore the expression of Bax in the retinal nervous tissue of early diabetic rats and the anti -aopotosis effect of TangWangMing . Methods:Diabetic mellitus in rats was induced by alloxan and divided into 2 groups, one group without treatment, the other group treated with TangWangMing, and normal rats used as control group. All rats were killed after 180 days , Expression of Bax in the retinal nervous tissue was studied by immunohistochemistry and the apoptotic retinal neural cells stained by the method of TUNEL were observed with light microscope . Results:Compared with control group, the intensity of Bax protein and apoptosis in the retinal neural cells were increased significantly in diabetic group without treatment. These changes in TangWangMing group werebetter than that of model group. Conclusion: TangWangMiug can suppress the expression of Bax and restrain the apoptosis of retinal neural cells. It may prevent the development of the early diabetic retinopathy and be useful in its treatment.
