首乌益智灵对血管性痴呆模型大鼠海马区神经元的影响

摘    要:目的:观察首乌益智灵对血管性痴呆(VD)模型大鼠脑组织海马区神经元细胞形态、海马区钙结合蛋白Calbindin-D-28k阳性神经元表达的影响。方法:用改良Pulsinelli四血管阻断(4-VO)法制造血管性痴呆大鼠模型,设首乌益智灵组、脑复康组、模型组、假手术组。分别灌胃20d后处死大鼠,取脑组织制作切片,HE和免疫组化染色后观察。结果:首乌益智灵组大鼠海马CA1区神经元排列基本规则,细胞轻度脱失,部分胞核染色变深;钙结合蛋白阳性神经元百分率及平均光密度均高于模型组和脑复康(P<0.05,P<0.01)。结论:首乌益智灵具有保护血管性痴呆模型大鼠海马区神经元、促进血管性痴呆大鼠钙结合蛋白阳性神经元表达的作用。

Abstract:Objective: To study the effect of Shou Wu Yi Zhi Ling on the rats with vascular dementia (VD) by observing the changes of the shape of neuron in the hippocamp, and the expression of Calbindin-D-28k in the area. Methods: The rat VD model was created with the reformed Pulsinelli 4-VO method, and rats were randomly divided into 4 groups including Shou Wu Yi Zhi Ling group, Nao Fu Kang group, Model group and Sham operation group. Rats in the two treatment group were feed by lavage for 20 days before executed. The collected brain tissues were processed into slice and dyed with HE and immunohistochamical method. Results: In the Shou Wu Yi Zhi Ling group, the neuron arrangement in CAI area of the hippocamp is regular just with mild cell loss and some darken dyeing nucleus; the rate of the expression of Calbindin-D-28k in the positive neurons and the average optical were higher than that in Model and Nao Fu Kang groups(P<0.05,P<0.01).Conclusion: Shou Wu Yi Zhi Ling is effective in protecting the neuron in the hippocamp of the rats with VD, and promoting the expression of Calbindin-D-28k in the positive neurons.
