| 摘 要： | 目的:研究神经生长因子(NGF)对山羊骨髓间充质干细胞(BMSCs)向成骨细胞分化的调节作用. 方法:体外培养山羊BMSCs,流式细胞仪鉴定表面标志物;将第3 代BMSCs分为空白对照组、成骨诱导对照组、NGF组和实验组,检测各组碱性磷酸酶(ALP)活性和骨钙素(OC)水平、应用von Kossa染色的方法比较各组细胞成骨性能. 结果:流式细胞术测得各组细胞均呈CD90、CD105强阳性表达,CD34、CD45阴性.实验组ALP和OC表达明显高于其它3 组,差异有显著性意义(P<0.05).成骨诱导对照组细胞von Kossa染色阳性,可见黑色钙化结节,实验组较成骨诱导对照组钙化结节数目明显增多,面积增大.而NGF组各项指标与空白对照组无明显差异.结论:单纯使用NGF并不能诱导山羊BMSCs向成骨细胞转化,但NGF对山羊BMSCs向成骨细胞分化过程具有明显的促进作用. |
