RECK和MMP-2在大肠癌中的表达及其相互关系

目的研究RECK和MMP-2在大肠癌及正常大肠组织中的表达，探讨其在大肠癌的发生、发展和浸润转移中的作用。方法应用免疫组织化学法（SABC）检测50例大肠癌组织和20例正常大肠黏膜组织中RECK和MMP-2的表达。结果RECK在大肠癌组织中的阳性表达率为64％，明显低于在正常大肠黏膜组织中的表达（100％）（P〈0．05），并随肿瘤病理分化程度的降低、临床Dukes分期的提高、淋巴转移的产生而降低（P〈0．05）。MMP一2在大肠癌组织中的阳性表达率为72％，明显高于在正常大肠黏膜组织中的表达（15％）（P〈0．05），并随临床Dukes分期的提高、淋巴转移的产生而增高（P〈0．05）。大肠癌组织中RECK与MMP-2的表达呈负相关（r=-0．642，P〈0．05）。结论大肠癌组织中RECK低表达，MMP-2高表达。RECK可能在大肠癌的发生和浸润转移过程中起重要作用，有望成为预测大肠癌转移与预后的重要参考指标。

Expression of RECK and MMP-2 and their relationship in colorectal carcinoma

Objective To study the expression of reversion-inducing cysteine-rich protein with Kazal motifs （RECK） and matrix metalloproteinase-2 （MMP-2） in colorectal cancer and normal colorectal mucosa, and to probe their roles on invasion and metabasis in colorectal carcinoma. Methods Immunochemical staining was used to detect the expression of RECK and MMP-2 protein in 50 cases of colorectal carcinoma and 20 cases of normal colorectal mucosa. Results The expression rate of RECK in colorectal carcinoma was significantly lower （64%） than that in normal color- ectal mucosa （ 100% ） （ P 〈 0.05 ）, and reduced with the decreased histological differentiation, the increased Dukes stages and occurrence of lymphatic （P 〈 0.05）. The expression rate of MMP-2 in colorectal carcinoma was significantly higher （72%） than that in normal colorectal mucosa （15%） （P 〈 0.05 ）, and elevated with the increased Dukes stages and occurrence of lymphatic （P 〈 0.05）. The expression of RECK was negatively correlated with the expression of MMP-2 （r =-0. 642, P 〈 0.05）. Conclusion RECK is lowly expressed in colorectal carcinoma, but MMP-2 is highly expressed. RECK and MMP-2 may play important roles in the infiltration and metastasis in colorectal carcinoma. Detecton of RECK may be used in prognosis judgement of colorectal carcinoma.
