摘    要:选择Ⅱ级Wistar大鼠,随机分为三组,自妊娠第14天开始皮下注射生理盐水、一氧化氮合成酶抑制剂亚硝基左旋精氨酸甲酯(L—NAME)或L—NAME联合一氧化氮供体硝普钠,直到分娩,观察抑制一氧化氮(NO)合成对孕鼠血压和重要器官的影响。结果L—NAME使孕鼠血压升高、尿蛋白增加,并导致妊娠高血压综合征样的肝肾功能损害及血液变化;硝普钠可部分对抗L—NAME的上述效应。提示妊娠期NO合成异常可能是导致妊娠高血压综合征发病的重要因素。

EFFECT OF NITRIC OXIDE SYNTHESIS INHIBITION ON MATERNAL ORGANS IN THE PREGNANT RATS
Abstract:Pregnancy-induced hypertension is associated with hypertension, proteinurea and injury of important organs .Our purpose was to determine the effects of nitric oxide synthase inhibition on maternal health in the last third of pregnancy. Wistar rats were chosen and divided into three groups randomly. Saline solution, an inhibitor of nitric oxide synthese, L-nitro-arginine methyl easer singly or associated with a nitrc oxide donor, nitroprusside sodium, was infused subcutaneously to rats from day 14 of gestation. Systolic blood pressure was recorded on day 14, 15, 17and 20 of gestation respectively. Maternal blood chemistry studies and urine protein were determined. Systolic blood pressures in rats infused with L-nitro-arginine methy1 easter were significantly elevated compared with controls (P<0.001); Maternal blood and urine chemistry studies were affected by L-nitro-arginine nethy1 easter, serum GPT(P<0.001),BUN(<0.001) ,CRE(P<0.05),UA(P<0.05), hematocrit (P<0.001) and urine protein (P<0.001) were increased significantly while blood platelet count decreased significantly(P<0.001). Except that serun GPT, hematocrit and platelet count were prevented partially ,others were prevented completely by coadministration of nitroprusside sodium. Infusion of an inhibitor of nitric oxide synthesis during pregnancy causes hypertension, proteinurea and pregnancy-induced hypertension-like alternation of maternal blood chemistry, and indicate that a decrease in nitric oxide syntheses may be one of the important factors responsible for pregnancy-induced hypertension.
