Survivin siRNA抑制人腺样囊性癌ACC-2细胞移植瘤血管生成

摘    要:目的 观察siRNA抑制生存素survivin基因对裸鼠皮下人腺样囊性癌ACC-2细胞移植瘤血管生成的影响.方法 裸鼠15只完全随机分为3组(n=5):①干扰质粒组,皮下接种survivin RNA干扰质粒转染的ACC-2细胞;②瘤体注射组,皮下接种正常ACC-2细胞15 d成瘤后,将针对survivin的siRNA真核表达载体脂质体法作瘤体内及瘤周注射;③肿瘤细胞组,皮下接种正常ACC-2细胞.25 d后处死全部裸鼠,剥离瘤体,测量瘤体体积,RT-PCR检测瘤体组织VEGFmRNA表达,HE染色和免疫组化染色法观察VEGF阳性染色及其微血管参数.结果 干扰质粒组和瘤体注射组VEGF电泳条带的相对光密度比值(0.698±0.023、0.612±0.038)及总体微血管密度(4.98.±1.43)、(5.08±1.29)个]、有腔微血管密度(2.98.±1.05)、(3.10.±1.23)个]、有腔血管周长(68.60.±15.98)、(70.12±14.19)μm]及管腔面积(4 119.14±698.12)、(4339.67±599.76)μm2]较肿瘤细胞组VEGF电泳条带的相对光密度比值(0.912 ±0.039)及总体微血管密度(8.90.±1.36)个]、有腔微血管密度(5.89.±1.68)个]、有腔血管周长(129.48.±25.68)μm]及管腔面积(6952.74.±1 298.54)μm2]低(P<0.05),而干扰质粒组和瘤体注射组无明显差别(P>0.05).结论 survivin siRNA能够抑制腺样囊性癌瘤体生长,并在体内通过抑制腺样囊性癌VEGF表达有效减少肿瘤血管生成.
