强脉冲光对成纤维细胞及血管内皮细胞增殖活性及血管内皮细胞生长因子分泌水平的影响

摘    要:目的:研究强脉冲光照射对皮肤成纤维细胞、血管内皮细胞的生长增殖状态及两种细胞中血管内皮细胞生长因子(VEGF)分泌水平的影响。方法:分离培养原代成纤维细胞和复苏血管内皮细胞株(ECV034),分别采用波长为590~1200nm和520~1200nm的能量密度为29J/cm2强脉冲光照射上述培养细胞。在照射结束后1、12、24及48h,以四甲基偶氮唑蓝(MTT)法测定细胞增殖活性情况,以酶联免疫吸附试验(ELISA)检测两种细胞培养上清液中VEGF的分泌情况。结果:成纤维细胞经强脉冲光照射24h及48h后,细胞增殖活性增强(P<0.05),而血管内皮细胞增殖活性无明显改变。强脉冲光照射对成纤维细胞及血管内皮细胞VEGF的分泌无影响。结论:强脉冲光能增强成纤维细胞增殖活性,但不影响成纤维细胞及血管内皮细胞VEGF的分泌。

Abstract:Objective: To investigate the effects of intense pulse light (IPL) on the cell proliferation and secretion of vascular endothelial growth factor (VEGF) in human fibroblasts and vascular endothelial cells. Methods: The infant foreskin fibroblasts and vascular endothelial cell line(ECV034) were cultured and treated with different wave length and designed dose of IPL irradiation (590nm, 520nm, and 29 J/cm2). By culturing 1, 12, 24 and 48 hours after the irradiation, the morphology change was observed by light microscopy, the cell proliferation activity was measured by MTT assay and VEGF secretion was determined by ELISA. Results: Compared with sham-irradiation, no obvious morphology change was observed for either cultured fibroblasts or vascular endothelial cells (ECV034). The cell growth or the cellular activity was increased (P < 0.05) in fibroblasts 24 h and 48 h after IPL irradiation. But the IPL irradiation showed no effect on VEGF level in fibroblasts at any time point after irradiation. For ECV034 cells there was no noticeable effect on cell morphology, cellular activity and VEGF secretion. Conclusions: IPL irradiation can induce the cell proliferation in cultured primary fibroblasts but no such effect on vascular cell line. IPL has no regulation on VEGF secretion for either cultured fibroblasts or ECV034.
