参芪扶正注射液延缓CCI大鼠神经病理性疼痛的产生

摘    要:目的:探讨早期给予参芪扶正注射液是否可以抑制坐骨神经慢性缩窄性损伤(CCI)大鼠神经病理性疼痛的产生.方法:SD大鼠32只,随机分为4组(n=8):①假手术组(sham组);②对照组(CCI组),即单纯行CCI手术组;③CCI+生理盐水组(CCI+saline组),于CCI术后立即腹腔注射生理盐水25mL/kg,连续7d,1次/d;④CCI+参附组(CCI+ShenFu组),于CCI术后即刻腹腔注射参芪扶正注射液,25mL/kg,连续7d,1次/d.于术前2d,术后1,3,7,10,14d测定各组大鼠热缩足反射潜伏期(TWL)、机械缩足反射阈值(MWT).结果:与Sham组比较,CCI组和CCI+Saline组术后各日TWL,MWT明显降低(P〈0.01),其MWT从术后第1日即开始下降,第7日降至最低值(为基础值的19.7%和18.1%,与基础值比较均P〈0.01);其TWT从术后第1日即开始缩短,第3日即缩短至最低值(为基础值的58.4%和60.2%,与基础值比较均P〈0.01),CCI组和CCI+Saline组两组间差异无统计学意义.与CCI,CCI+Saline组相比,CCI+ShenFu组在术后第1,3,7日的MWT降低较为缓和(P〈0.01),但自术后第10日始,各组间差异无统计学意义;CCI+ShenFu组在术后第1日TWL缩短较为缓和(P〈0.01),但自术后第7日始,各组间差异无统计学意义.结论:早期给予参芪扶正注射液可以轻度延缓CCI大鼠机械性触诱发痛的形成,抑制术后第1日的热痛觉过敏.

Abstract:AIM: To investigate whether Shenfu Injection, an intravenous injection made from traditional Chinese medicinal herbs, could postpone the production of neuropathic pain in CCI rats. METHODS: Thirty-two SD rats were divided into 4 groups randomly: (1)CCI group in which the rats underwent the CCI (the sciatic nerve chronic constriction injury ) procedure, (2)CCI + saline group in which the rats were given saline injection (25 mL/kg) once daily for 7 consecutive days immediately after the CCI procedure, (3)CCI + Shenfu group in which the rats were given Shenfu injection ( 25 mL/kg) once daily for 7 consecutive days immediately after the CCI procedure, and (4)Sham group in which the rats underwent the same procedure as CCI group but without ligature of the sciatic nerve. At -2, 1,3, 7, 10 and 14 d after the surgery, we examined the time course of hyperalgesia including thermal paw withdrawal latency and Van Frey withdrawal threshold. RESULTS: Compared with those in Sham group, the scores of thermal paw withdrawal latency (TWL) and Von Frey withdrawal threshold (MWT) in CCI group and CCI + saline group decreased significantly after surgery(P 〈0.01 ). The scores of MWT in these two groups declined from the first day and even- tually reached the bottom at the 7th day after surgery. The scores of TWL in these two groups were significantly lower than the scores in Sham group from the first day after surgery, and reached the bottom at 3rd day after surgery. There was no significant difference in TWL and MWT between CCI group and CCI + saline group. At 1, 3 and 7 d after surgery, the scores of MWT in CCI + Shenfu group were significantly higher than those in CCI and CCI + saline group(P 〈0.05). However, There was no significant difference in TWL and MWT since the 10th after surgery. Compared with those in CCI and CCI + saline group, the scores of TWL in CCI + Shenfu group were significantly higher than those in CCI and CCI + saline group only on the 1
