黄芪葛根配伍对糖尿病大鼠脂肪组织AMPK信号通路的影响

摘    要:目的探讨黄芪与葛根配伍对糖尿病脂肪中腺苷酸活化蛋白激酶(AMPK)信号通路的作用,明确其调控血糖血脂机制。方法将86只SPF级SD雄性大鼠按照随机数字表分成6组:正常组、模型组、金芪降糖组、黄芪组、葛根组、黄芪葛根配伍组,正常组11只,其余各组15只大鼠。一次性腹腔注射2%STZ(46 mg/kg)造模,同日各组分别予金芪降糖混悬液1.47 g/kg、黄芪2.7 g/kg、葛根1.35 g/kg、黄芪葛根汤4.05 g/kg灌胃,给药30 d。酶联免疫吸附测定(ELISA)检测脂联素(APN)、葡萄糖转运体4(GLUT-4),实时荧光定量聚合酶链式反应法(Real-time PCR)检测脂联素受体1(AdipoR1)、脂联素受体2(AdipoR2)、AMPK、GLUT-4、过氧化物酶增殖体激活受体-α(PPARα)、PPARγmRNA。结果与正常组比较,模型组大鼠APN、GLUT-4水平降低,AdipoR1、AdipoR2、AMPK、GLUT-4、PPARα、PPARγmRNA表达减少(P<0.05)。黄芪、黄芪与葛根配伍能升高模型大鼠APN、GLUT-4水平(P<0.05);黄芪、葛根及两药配伍能增加AdipoR1(葛根、黄芪与葛根配伍除外)、AdipoR2、AMPK、PPARα(葛根除外)、PPARγ、GLUT-4mRNA表达(P<0.05),黄芪促进AdipoR2、PPARαmRNA表达的作用优于两药合用,黄芪与葛根配伍调节AMPK、PPARγ、GLUT-4mRNA表达的作用优于单一用药(P<0.05)。结论黄芪、葛根及其配伍调节血糖血脂与调控糖尿病大鼠脂肪组织AMPK信号通路有关,黄芪葛根配伍调控血糖、血脂作用优于单一用药,可能与其促进APN、AMPK、GLUT-4、PPARγmRNA表达有关。

Abstract:Objective To observe the effect of Huangqi(Radix Astragali)-Gegen(Radix Pueraria Lobata)compatibility on AMP-activated protein kinase(AMPK)signaling pathway in adipose tissue of diabetic rats and its mechanism of reducing blood glucose and blood lipid.Methods Eighty-six SPF male SD rats were divided into 6 groups:normal group,model group,Jinqi Jiangtang group,Huangqi(Radix Astragali)group,Gegen(Radix Pueraria Lobata)group and Huangqi(Radix Astragali)-Gegen(Radix Pueraria Lobata)compatibility group by the random number table.There were 11 rats in normal group and 15 rats in the other groups.Except the normal group,2%STZ(46 mg/kg)was given to rats of the other groups by intraperitoneal injection to induce diabetes models.At the same time,the suspension of Jinqi Jiangtang Tablet with the dose of 1.47 g/kg was given to rats in control group.Huangqi(Radix Astragali)with the dose of 2.7 g/kg was given to rats in Huangqi(Radix Astragali)group.Gegen(Radix Pueraria Lobata)with the dose of 1.35 g/kg was given to Gegen(Radix Pueraria Lobata)group.Huangqi Gegen Decoction with the dose of 4.05 g/kg were given to Huangqi(Radix Astragali)-Gegen(Radix Pueraria Lobata)compatibility group.All of drugs were given for 30 days continuously.The contents of adiponectin(APN)and glucose transporter4(GLUT-4)were detected by enzyme-linked immunosorbent assay(ELISA).The relative expression of adiponectin receptor 1(AdipoR1),adiponectin receptor 2(AdipoR2),AMPK,GLUT-4,peroxisome proliferator-activated receptorα(PPARα)and PPARγmRNA were tested by Real-time PCR.Results Compared with those of the normal group,the contents of APN and GLUT-4 were lowered,while the relative expressions of AdipoR1,AdipoR2,AMPK,GLUT-4,PPARαand PPARγmRNA were reduced in the model group(P<0.05).Huangqi(Radix Astragali)and Huangqi(Radix Astragali)-Gegen(Radix Pueraria Lobata)compatibility could increase APN and GLUT-4 in model group(P<0.05).Huangqi(Radix Astragali),Gegen(Radix Pueraria Lobata)and their compatibility could increase the relative expression of AdipoR1except Gegen(Radix Pueraria Lobata)and Huangqi(Radix Astragali)-Gegen(Radix Pueraria Lobata)compatibility],AdipoR2,AMPK,GLUT-4,PPARαexcept Gegen(Radix Pueraria Lobata)],PPARγmRNA in model group(P<0.05).The effect of Huangqi(Radix Astragali)on AdipoR2 and PPARαmRNA expression was better than Huangqi(Radix Astragali)-Gegen(Radix Pueraria Lobata)compatibility.The effect of Huangqi(Radix Astragali)-Gegen(Radix Pueraria Lobata)compatibility on regulating AMPK PPARγand GLUT-4 mRNA expression was better than single drug.Conclusion Huangqi(Radix Astragali),Gegen(Radix Pueraria Lobata)and their compatibility to reduce blood glucose and blood lipid were related to regulating AMPK signal pathway in adipose tissue of diabetic rats.The effect of Huangqi(Radix Astragali)-Gegen(Radix Pueraria Lobata)compatibility in regulating blood glucose and blood lipid was better than single drug.It may be related to Huangqi(Radix Astragali)-Gegen(Radix Pueraria Lobata)compatibility to promote the expressions of APN,AMPK,GLUT-4 and PPARγmRNA.
