鸢尾黄素对感染后咳嗽豚鼠的抗气道炎症作用

摘    要:目的探讨射干提取物中鸢尾黄素对感染后豚鼠咳嗽模型的抗气道炎症作用。方法采用呼吸道合胞病毒(RSV)诱导及烟熏法建立感染后咳嗽豚鼠模型,采用全自动模块式血液体液分析仪测定血液及肺泡灌洗液中炎性细胞计数,采用ELISA试剂盒检测血清和肺泡灌洗液中IL-4和IFN-γ含量,并计算比值。结果豚鼠模型组病毒学检测和组织病理学结果显示RSV病毒感染成功,且未造成实质性肺炎;模型组5 min内咳嗽次数显著高于空白组,成功复制感染后咳嗽模型。给药前,造模对血液中炎性细胞计数无改变,但升高肺泡灌洗液中白细胞总数和中性粒细胞计数,降低血清中IFN-γ的含量,降低肺泡灌洗液中IL-4含量,升高IFN-γ的含量;给药后,鸢尾黄素可显著升高肺细胞灌洗液中INF-γ的含量,降低IL-4/IFN-γ的比值。结论射干中鸢尾黄素对RSV感染后咳嗽豚鼠有较好的抗气道炎症作用。

Abstract:Objective To investigate the application and anti-inflammatory effects of tectorigenin in guninea pigs with postinfectious cough by respiratory syncytial virus(RSV).Methods The model of guinea pigs with postinfectious cough established by smoking and infecting with RSV.The total leukocyte count and the number of other inflammatory cells in alveolar lavage fluid were detected by blood body fluid analyzer.The contents of cytokines(IL-4 and IFN-γ)in BALF(bronchoalveolar lavage fluid)and serum were detected by ELISA kit method,and the ratio of them was calculated.Result It was successful that in the model group guinea pig was infected by RSV,which was showed by the result of virological testing and histopathological examination.Furthermore,this method did not lead to the structural damage of the guinea pig lung.The cough frequency in the model group was significantly higher than that in the blank group within 5 min.This model method did not change the number of inflammatory cell.In BALF,it increased the number of white cell and neutrophil and the content of IFN-γand decreased the content of IL-4.In serum,it decreased the content of IFN-γ.After dosing,tectorigenin could significantly increase the content of IFN-γ,and decrease the ratio of IL-4 and IFN-γ.Conclusion The tectorigenin extracted from Shegan(Belamcanda chinecsis)had the anti-inflammation effect to guinea pigs with postinfectious cough by RSV.
