常山水提醇沉物对EC9706细胞周期、凋亡、能量代谢的影响

目的从细胞增殖、凋亡、细胞周期、能量代谢角度,研究常山水提醇沉物抗食管癌细胞EC9706的作用及机制。方法用MTT法检测EC9706细胞活性,流式细胞术检测药物对食管癌EC9706细胞凋亡、周期的影响,能量代谢检测系统检测药物对EC9706细胞能量代谢的影响。结果不同浓度常山水提醇沉物作用48 h后能有效抑制EC9706细胞增殖,呈剂量依赖性(P<0.01),可以有效促进细胞凋亡(P<0.05),将EC9706细胞阻滞于S期和G2/M期(P<0.05),并且明显抑制细胞糖酵解及线粒体代谢能力(P<0.01)。结论以上结果表明,常山水提醇沉物可能通过干预细胞周期、凋亡及能量代谢抑制食管癌EC9706细胞增殖。

Effects of Water Extraction-Alcohol Precipitation Extract of Changshan(Dichroa)on Cell Cycle,Apoptosis and Energy Metabolism of Esophageal Carcinoma EC9706

Objective From the perspective of cell proliferation,apoptosis,cell cycle and energy metabolism,to study the mechanism of treating esophageal cancer with water extraction-alcohol precipitation extract of Changshan(Dichroa).Methods MTT was used to detect cell proliferation,flow cytometry was used to detect its effect on apoptosis and cycle of esophageal cancer EC9706 cells,and energy metabolism was used to detect the effect of drugs on energy metabolism of EC9706 cells.Results After 48 h treatment with different concentrations,the extracts could effectively inhibit the proliferation of EC9706 cells in a dose-dependent manner(P<0.01),increase the percentage of EC9706 cells at S phase and G2/M phase(P<0.05),promote cell apoptosis(P<0.01),and significantly inhibit cell glycolysis and mitochondrial metabolism(P<0.01),but there was no significant difference between low and high concentrations.Conclusion These results suggest that water extraction-alcohol precipitation extract of Changshan(Dichroa)may inhibit the proliferation of esophageal carcinoma EC9706 cells by intervening cycle,apoptosis and energy metabolism.
