基于网络药理学研究巴戟天抗抑郁的分子作用机制

摘    要:目的运用网络药理学的方法探讨巴戟天治疗抑郁症的分子作用机制。方法通过中药系统药理学数据库与分析平台(TCMSP数据库)筛选出巴戟天的活性成分和对应的作用靶点;利用GeneCard、OMIM等数据库筛选出抑郁症的潜在靶点;采用Cytoscape 3.6.1软件构建药物-成分-疾病-靶点的可视化网络;以STRING数据库对关建靶点构建蛋白互作网络分析;最后进行基因本体论(GO)富集分析、京都基因与基因组百科全书(KEGG)通路富集分析。结果从巴戟天中筛选出有效成分20个,得到可作用于抑郁症的靶点22个。GO分析富集得到65个生物学功能,KEGG通路分析富集得到48条信号通路。结论巴戟天通过多靶点、多通路发挥对抑郁症的治疗作用,其作用机制可能与胆碱能突触、神经活性配体-受体相互作用、甲状腺激素信号通路、PI3K-Akt信号通路、雌激素信号通路等相关。

Abstract:Objective To explore the molecular mechanism of Bajitian(Morindae Officinalis Radix)for treatment of depression based on network pharmacology.Methods TCM Systems Pharmacology Database and Analysis Platform were used to screen out the active constituents and the targets of the constituents from Bajitian(Morindae Officinalis Radix).All targets of depression were collected in OMIM,TTD and other databases.Cytoscape 3.6.1 software was used to construct a visual network topological relationship map of the drug-constituent-disease-target.And the analysis of protein-protein interaction network was carried out based on string database.Finally,the enrichment analysis of gene ontology(GO)biological process and KEGG pathway enrichment analysis were carried out.Result Twenty active constituents of Bajitian(Morindae Officinalis Radix)were screened out and 22 related targets were obtained which could act on depression.In the GO functional enrichment analysis,there were 65 GO terms.The KEGG pathway enrichment analysis contained 48 pathways.Conclusion The therapeutic effect of Bajitian(Morindae Officinalis Radix)on depression may be realized through a multi-target and multi-pathway method,and cholinergic synapse,neuroactive ligand-receptor interaction,thyroid hormone signaling pathway,PI3 K-Akt signaling pathway and estrogen signaling pathway may play an important role in depression treated by Bajitian(Morindae Officinalis Radix).
