超声辅助低共熔溶剂法提取黄芩中3种黄酮类成分的工艺研究

摘    要:目的以氯化胆碱-乳酸组成的低共熔溶剂为提取介质,采用超声辅助的方法提取黄芩中的黄芩苷、汉黄芩苷和黄芩素3种黄酮成分。方法单因素实验法考察了超声时间、含水量以及液料比对3种黄酮成分总得率的影响,并运用星点设计-响应面法进行了优化。结果黄芩中3种黄酮成分提取的最佳条件为超声时间15 min、含水量18%、液料比23 mL·g-1,该条件下3种黄酮成分的总得率为224 mg·g-1,与模型的理论值(230 mg·g-1)的相对误差为2.61%,说明建立的模型有效、可靠。结论超声辅助低共熔溶剂法提取具有得率高、速度快、安全环保等优点,该研究为黄芩中3种黄酮成分的提取提供了一种新方法,同时也为天然产物中活性组分的提取提供了参考。

Abstract:Objective The deep eutectic solvents(DESs)consisting of chloride and lactic acid was used as solvent to extract baicalin,wogonoside and baicalein with ultrasound-assisted method from Huangqin(Scutellaria baicalensis Georgi).Methods The effects of ultrasound time,water ratio and the ratio of liquid to solid on the total yield of flavonoids were researched by single factor experiment.On this basis,the central composite design-response surface methodology was used to optimize the extraction process.Results The results showed that optimal extraction conditions of flavonoids from Huangqin(Scutellaria baicalensis Georgi)were ultrasound time 15 min,water ratio 18%and the ratio of liquid to solid 23 mL·g-1.The total yield of flavonoids could be up to 224 mg·g-1,and the relative error was 2.61%compared to theoretical value(230 mg·g-1),which indicated the feasible model fitted well with the experimental data.Conclusion The ultrasound-DESs method has the advantages of high yield,fast speed,safety and environmental protection.This study not only provides a new method for the extraction of three flavonoids from Huangqin(Scutellaria baicalensis Georgi),but also provides a reference for the extraction of active components from natural products.
