氨基胍对糖尿病大鼠肾脏中AGEs和LPO的影响

摘    要:目的探讨氨基胍(AG)对糖尿病大鼠肾脏中糖基化终末产物(AGEs)和脂质过氧化物(LPO)的影响。方法将36只雄性SD大鼠随机分为正常对照组(N组)、模型组(DM组)和AG组,用链脲佐菌素制作糖尿病大鼠模型。AG组给予AG治疗8周。测定各组大鼠肾质量/体质量、血尿素氮(BUN)、血肌酐(SCr)2、4 h尿蛋白定量、肾小球AGEs(GTE-AGEs)、血清和尿中抗CML-AGEs及LMN-AGEs、肾小球和血清LPO含量,观察肾脏病理改变。结果DM组大鼠血清BUN、SCr、血清AGEs、血清LPO、GTE-AGEs、GTE-LPO、肾质量/体质量、24 h尿蛋白定量均显著高于N组,AG组血清BUN、血清SCr、血清CML-AGEs、血清LPO、GTE-AGEs、GTE-LPO、肾质量/体质量、24 h尿蛋白定量均显著低于N组,尿中AGEs、血清CML-AGE含量显著高于DM组和N组;AG可显著减轻肾脏病理学改变。结论氨基胍可通过减少AGEs在糖尿病大鼠肾脏的蓄积,降低尿蛋白排泄,减少氧化应激水平及其他间接作用,发挥其对糖尿病肾病的防治作用。

Abstract:Objective It is to explore the effect of aminoguanidine(AG) on the advanced glycosylation end products(AGEs) and lipid peroxides in kidneys in diabetic rats.Methods 36 male SD rats were randomly divided into 3 groups: normal control group(group N),model group(group DM) and group AG.Diabetes models were induced by injection of streptozocin.Group AG was treated with AG for 8 weeks. Kidney weight/body weight,serum Cr,BUN,AGEs,LPO,glomerular tissue extracts AGEs(GTE-AGEs),glomerular tissue extracts(GTE-LPO),glomerular mesangial volume,24 h urine protein and urine AGEs were measured in each group.The pathological changes of kidney were observed.Results After treatment,the levels of serum Cr,BUN,AGEs,LPO,GTE-AGEs and GTE-LPO,glomerular mesangial volume,kidney weight/body weight,24 h urine protein and urine AGEs were all significantly higher in group DM than that in group N,but the indexes above in group AG were all significantly lower than that in group N,the contents of urine AGEs and serum CML-AGE in group AG were obviously higher that in group DM and group N.AG could improve the pathologic changes of kidney in the rats.Conclusion AG may exert its renoprotective effect through inhibiting the accumulation of renal AGEs,decrease urinary albumin excretion,decrease deposition of oxidative stress and other indirect ways.
