Abstract:Objective To investigate the expression of microRNA (miRNA/miR)-146 in the serum of patients with bronchiectasis and bronchial asthma and its influence on lung function. Methods 156 patients with bronchiectasis and bronchial asthma admitted in the Third Hospital of Xingtai from January 2017 to August 2020 were the subjects of the study, and 150 healthy subjects during the same period were used as the control group. The real-time quantitative polymerase chain reaction method was used to detect the expression level of miR-146, the enzyme-linked immunosorbent assay was used to determine the level of serum inflammatory factors and the lung function was used to test their lung function indicators.Pearson correlation was used to analyze the relationship between miR-146 expression level and lung function. Results The lung function indexes of the control group and the observation group were FVC (3.34±0.92L vs 2.68±0.84L), FEV1 (2.03±0.26L vs 1.65±0.23L), FEV1 /FVC (82.67±7.14% vs 65.03±6.96%), MMEF75/25 (1.28±0.23L/s vs 0.57±0.12L/s) and PEF (545.28±15.89L/min vs 140.57±13.65L/min), respectively. Compared with the two groups, the lung function indexes FVC, FEV1, FEV1 /FVC, MMEF75/25 and PEF in the observation group were significantly decreased ,the differences were statistically significant (t=6.558~239.284, all P<0.01). Serum miR-146a (2.51±0.68), miR-146b mRNA (1.68±0.49), the expression levels of IL-4 (325.41±32.78ng/L) and INF-γ (232.12±18.48ng/L) were significantly higher than those of the control group (0.98±0.41, 1.01±0.36, 228.69±26.15ng/L, 202.25±17.36ng/L), the differences were statistically significant (t= 13.667~28.587, all P < 0.001). Pearson correlation analysis showed that the expression levels of miRNA-146a and miRNA -146b in patients with bronchiectasis and bronchial asthma were significantly correlated with FVC,FEV1, FEV1 /FVC and MMEF75/25. There was a significant negative correlation between PEF and other lung function indexes (r =-0.687~0.458, P<0.05), and a significant positive correlation between PEF and the expression levels of inflammatory cytokines IL-4 and INF-γ (r=0.698~0.745, all P<0.001). Conclusion MiR-146 was highly expressed in the serum of bronchiectasis with bronchial asthma, and the expression level of miR-146 was significantly negatively correlated with lung function, which has certain value for lung function assessment.
