双链RNA对人鼻息肉上皮细胞中基质金属蛋白酶mRNA表达的影响

摘    要:目的 病毒感染经常使慢性鼻-鼻窦炎(chronic rhinosinusitis,CRS)病情加重.病毒感染时,基质金属蛋白酶(matrix metalloproteinases,MMPs)调节机制的破坏是导致病情加重的机制之一.本研究试图更好地了解病毒感染后鼻上皮细胞MMPs的表达.方法 取鼻内镜手术时CRS患者的鼻息肉组织,分离培养鼻上皮细胞,以合成的双链RNA类似物--多聚次黄嘌呤胞嘧啶核苷酸[polyinosinic-polycytidylic acid,poly(I:C)]刺激原代鼻上皮细胞后,检测鼻上皮细胞中MMP-2、MMP-9和组织金属蛋白酶抑制物-1(tissue inhibitor of metalloproteinase-1,TIMP-1)mRNA的相对表达量.结果 poly(I:C)刺激后,鼻上皮细胞中MMP-9 mRNA的相对表达量((x-)±s,以下同)较同一患者未刺激的细胞增高(22.61±5.47)倍,差异有统计学意义(Z=-2.52,P=0.012).poly(I:C)刺激后,鼻上皮细胞中MMP-2和TIMP-1 mRNA的相对表达量没有明显改变.结论 双链RNA刺激离体培养的人鼻上皮细胞后,MMP-9 mRNA表达显著增高,且不受TIMP-1的调节.由此可能导致CRS患者病情加重.

Abstract:Objective Chronic rhinosinusitis was often exacerbated by viral infection.A disruption of the mechanisms that regulate the activity of matrix metallopmteinases(MMPs)during viral infection was one possible mechanism responsible for the cxacerbation.The purpose of study was to achieve a better understanding of MMP expression in hasal epithelial cells after viral infection.Methotis Human nasal epithelial cells were isolated from nasal polyp specimens obtained during endoscopic endonasal surgery in chronic rhinosinusitis patients.The expression of MMP-2,MMP-9,and tissue inhibitor of mctallopmteinase (TIMP).1 mRNA in primary human nasal polyp epithelial cells after double stranded RNA(ds RNA) stimulation were investigated.Results Among the genes whose expression was evaluated,only expression of MMP-9 mRNA increased significantly after dsRNA stimulation(22.61±5.47 fold increase,Z=-2.52,P=0.012).Conclusions The significant up-regulation of MMP-9 mRNA,which Was not modulated by TIMP-1,was an additional siyrec of increased pmteolytic activity in virus-infected upper airways that might contribute to the exacerbation of chronic rhinosinusitis with nasal polyps.
