Alzheimer病和血管性痴呆患者载脂蛋白E基因多态性分析

探讨ApoE多态性与Alzheimer病（AD）和血管性痴呆（VD）的关系。方法：应用PCR—RFLP技术分析25例AD，30例VD及40例对照组人群的ApoE基因型。结果：与对照组相比较，AD和VD患者ε3频率降低（P＜0．05），ε4频率升高（P＜0．05），两组患者间各等位基因频率差异无统计学意义（P＜0．05）。结论：ApoE多态性与AD和VD的发病机制相关，其在这两种疾病中的作用相似。

An analysis of Apolipoprotein E polymorphism in patients with Alzhemier's disease and vascular dementia

Objective To detect the polymorphism of Apolipoprotein E (APOE) in patients with Alzgeimer's disease (AD) and vascular dementia (VD), find the possible APOE roles in the pathogenesis of these two dementias. Methods We collected 25 sampleS of AD and 30 samples of VD. groups,Conclusion ApoE Polymorphism hae a correlation with AD and VD. DNA from patients was extracted from the peripheral blood samples. ApoEgenotypes were investigated with the methods of polymerase chain reaction (PCR) and restriction fragment length polymorphism(RFLP). Results The frequency of ε3 allele were 72. 0%, 70. 0%, 86. 3% and ε4 allele were 22. 0%, 21. 7%, 7. 4% in AD, VD and contros,respeCtively. The decrease ε3 allele frequencies (P<0. 05) and increase ε4 allele frequencies (<0.05)were found in both AD and VD. groups, conclusion APCE pclymcrphism has a corration with AD and VDεd, ε4 allele may be protective and risk factor, respectively. It could have the similar mechanism in the pathogenesis of AD and VD.
