皮瓣缺血再灌注损伤过程髓过氧化酶及白细胞膜CD_18的变化

摘    要:目的观察大鼠岛状皮瓣在缺血再灌注损伤过程中髓过氧化酶 (MPO)含量及外周血白细胞膜CD18 的变化。方法应用大鼠腹部岛状皮瓣缺血再灌注损伤模型 ,检测皮瓣髓过氧化酶 (MPO)含量 ,测定外周血白细胞膜CD18 水平的变化并作白细胞计数。结果缺血8h及缺血再灌注1h白细胞膜CD18 水平与皮瓣MPO水平较对照组明显增高 (P<0.01) ,而缺血再灌注1h较缺血8h组又明显增高 (P<0.01)。缺血再灌注1h组外周血白细胞计数较对照组明显减少 (P<0.01)。结论大鼠岛状皮瓣缺血再灌注损伤过程中外周血白细胞膜CD18 水平增高 ,白细胞计数减少 ,皮瓣MPO水平增高。CD18 介导的白细胞粘附在皮瓣缺血再灌注损伤中起重要作用

Abstract:Objective To investigate the change of Leukocyte CD18 and myeloperoxidase content during rat island flap reperfusion injury.Methods we utilized an island flap model in rat to detect the level of peripheral Leukocyte CD18, the tissue contents of myeloperoxidase(MPO) in flaps and the Leukocyte count of the circulating blood.Results The level of leukocyte CD18 and MPO both increased significantly by 8 hours' ischemia and 1h reperfusion compared with those of the controls(P<0.01). The level of Leukocyte CD18 and MPO of the reperfusion 1 h group increased significantly compared with that of ischemia 8 hours group(P<0.01). The leukocyte count of the circulating blood was lower in the reperfusion 1h group than that in the control group(P<0.01). Conclusion The level of leukocyte CD18 and MPO increases and the leukocyte count decreases during reperfusion injury of rat island flaps.CD18_mediated leukocyte adherence plays an important role in reperfusion injury.
