摘    要:目的建立葛丹颗粒中葛根素含量的测定方法。方法采用超高效液相色谱法(UPLC)对方中君药葛根的主要活性成分葛根素进行含量测定,色谱条件为ACQUITY UPLC BEH C18(2.1 mm×50 mm,1.7μm)色谱柱,流动相:乙腈—0.1%磷酸(15∶85);流速:0.2 ml.min-1;柱温:30℃;检测波长:250 nm。结果葛根素在0.021 9~0.390 0 g.L-1范围内呈良好的线性关系,r=0.999 9,平均回收率为98.84%,RSD=0.71%。结论该法简单、快捷,适用于葛丹颗粒中葛根素的含量测定。

Abstract:Objective To establish a method for determination of puerarin in gedan granule. Methods The content of puerarin was de- termined by UPLC. The chromatographic conditions were as follows:ACQUITY UPLC BEH Cls (2.1 mm×50mm,1.7μm) used as analyrical column. The mobile phase consisted of a mixture of V (acetonitrile) : V ( ortho-phosphoric acid) = 15: 85 and the flow rate was 0.2 mL min - l, with column temperature at 30℃, and UV detection at 250 nm. Results The puerarin was linear in the range of 0. 021 9 0. 390 0 g L- 1, r = 0.999 9. The average recovery rate was 98.84% ( RSD = 0.71% ). Conclusion This method can be used to con- trol the quality of puerarin in gedan granule better.
