右旋糖酐-磁性层状复合氢氧化合物-氟尿嘧啶转运模型在正常大鼠体内分布的磁靶向性

摘    要: 目的 研究右旋糖酐-磁性层状复合氢氧化合物-氟尿嘧啶转运模型在大鼠体内药物分布的磁靶向性。方法 建立高效液相色谱内标法,测定给药后大鼠体内各组织的氟尿嘧啶分布浓度;用紫外分光光度法、原子吸收法及等离子体发射光谱法测定给药干预后大鼠各组织铁的分布浓度,通过普鲁士蓝铁染色直观评价铁的组织分布差异。结果 高效液相色谱内标法回收率高、精密度好;经腹腔注射给药后,背右侧及头部设立磁场部位的药物浓度明显高于其他远离磁场的组织部位,数值分布有统计学意义,证明三级复合模型具有理想的磁靶向运转效果。结论 在外加磁场的作用下,右旋糖酐-磁性层状复合氢氧化合物-氟尿嘧啶转运模型能在靶向部位明显富集,表现良好的磁靶向性。

Abstract:OBJECTIVE To observe the magnetic targeting drug bio-distribution of the transshipment model with a form of Dextran -Magnetic LDH- Fluorouracil in rats. METHODS A HPLC method was established to determine the concentration of 5-Fu in tissues. The ultraviolet spectro-photometry, flame atomic absorption spectrometer and the inductively coupled plasma were used to detect the iron concentration in mouse tissues, and Prussian blue iron stain was employed for observing the different of iron bio-distribution in tissues. RESULTS The established HPLC method had high recovery and a good precision. After administration via peritoneal injection,the magnetic field had been set up in right of the back and the head, and the drug concentration were significant higher than other parts which were far away from the magnetic field. Drug concentration distribution showed statistical significance. This ternary composite model had an ideal magnetic targeting transfer effect. CONCLUSION Under the influence of an external magnetic field, the Dextran -Magnetic LDH- Fluorouracil transshipment model could be significantly enriched in the target area to show a good magnetic targeting.
