摘    要: 目的 研究前列地尔(PGE1对兔内皮细胞缺氧复氧损伤后抗栓功能的影响。方法 采用体外培养的兔内皮细胞建立缺氧复氧损伤模型, 实验分正常组、模型组、加入不同浓度(0.05,0.20,0.40 μg·mL-1PGE1的给药组,将PGE1分别加入缺氧复氧液中调整至以上终浓度。测PGE1(0.05,0.20,0.40 μg·mL-1不同浓度组及模型组、对照组培养上清液中丙二醛(MDA含量、超氧化物歧化酶(SOD活力和一氧化氮(NO含量、细胞内血管假性血友病因子(vWF的表达。结果 与正常组比较,模型组丙二醛(MDA含量增加,SOD活性、NO含量下降, vWF表达下降(P<0. 05。PGE1各浓度组与模型组比较,PGE1各浓度组SOD活性及NO含量有所增加,丙二醛(MDA含量降低,vWF表达增强(P<0. 05。结论 PGE1对缺氧/复氧损伤后血管内皮细胞的抗栓功能有保护作用。

Abstract:OBJECTIVE To study the effect of alprostadil (PGE1 on antithrombotic function in rabbit endothelial cells injuried by hypoxia/reoxygenation(H/R.METHODS H/R injury model was established with the cultured endothelial cells from rabbits in vitro.The endothelial cells were divided into normal group, model group and treatment groups with different concentrations of PGE1(0.05, 0.20, 0.40 μg·mL-1. PGE1 was added to the liquid of H/R and adjusted to a final concentration mentioned above . Malondialdehyde (MDA content, the activity of superoxide dismutase (SOD, and the nitric oxide (NO content in the culture supernatants and the expression of von Willebrand factor (vWF in cells sampled from different group (normal, model and treatment groups were assayed.RESULTS Compared with normal group,MDA content in model group was increased, while the activity of SOD, NO content and the expression of VWF in model group were reduced(P<0.05. Compared with model group, the activity of SOD and NO content in treatment groups with different concentrations of PGE1were increased, while MDA contents in treatment groups were reduced, and the expression of vWF was enhanced(P<0.05. CONCLUSION PGE1 has protective effect on antithrombotic function in rabbit endothelial cells injuried by H/R.
