摘    要: 目的 采用与介孔材料复合的方式提高难溶性药物水飞蓟宾的溶出速率。方法 通过X射线衍射、氮气吸附-脱附曲线、差示扫描量热分析及透射电镜观察水飞蓟宾负载到SBA-15的表面情况。气相色谱法检测水飞蓟宾负载后的丙酮残留情况。溶出速率实验研究水飞蓟宾负载到SBA-15的表面效果。结果 X射线衍射、氮气吸附-脱附曲线与差示扫描量热分析表明水飞蓟宾以非晶的形式负载在SBA-15的孔道内表面,负载前后的透射电镜图片显示无显著差异。气相色谱检测法表明丙酮残留符合要求。溶出速率实验表明负载后水飞蓟宾的溶出速率显著提高。结论 本实验为提高难溶性药物的溶出速率提供了一条新途径。

Abstract:OBJECTIVE To improve the dissolution rate of silybin by loading it into SBA-15 mesoporous silica. METHODS The adsorption of silybin on the surface of SBA-15 was observed by X-ray diffraction, N2 adsorption -desorption isotherms, differential scanning calorimeter experiments and TEM. A GC method was established for the determination of residual acetone in SBA-15/SB . The effect of silybin attachment on the surface of SBA-15 was investigated in dissolution experiment. RESULTS X-ray diffraction, N2 adsorption-desorption isotherms and differential scanning calorimeter experiments showed that amorphous silybin was densely and uniformly loaded on the surface of the pore in SBA-15. There was no difference between SC and SBA-15/SB in the image of TEM. The quantity of residual acetone in SBA-15/SB met the requirement as showen by GC. The dissolution experiment indicated that the dissolution rate of composite was increased significantly compared to crystalline silybin. CONCLUSION This study provides a new way to improve the dissolution rate of poor water soluble drugs.
