摘    要:目的 :研究单核细胞与血管平滑肌细胞 (VSMCs)之间的相互作用对基质金属蛋白酶 - 1(MMP- 1)分泌的影响。  方法 :将人类单核细胞与 VSMCs联合及隔离培养 ,用酶联免疫荧光法测定 MMP- 1水平 ,并用荧光标记胶原蛋白消化法测定 MMP- 1的活性。  结果 :单核细胞与 VSMCs联合培养可明显增加 MMP- 1的水平和活性 ,当两种细胞之比为 1.0时达高峰 ;细胞间的直接接触及细胞因子可介导 MMP- 1的产生。  结论 :单核细胞与 VSMCs之间的相互作用可促使 MMP- 1的产生及活性增加 ,可能在动脉粥样硬化斑块的破裂中起重要作用。

Abstract:Objective:To investigate the secration of matrix metallproteinase 1 (MMP 1) by a cell to cell interaction between monocytes and vascular smooth muscle cells (VSMCs). Methods:Human VSMCs and human monocytoid cells (THP 1 cells) were cocultured.MMP 1 levels were measured by enzyme linked immunosorbent assay.MMP 1 activity was determined by fluorescence labeled collagen digestion. Results:The coculture of THP 1 cells and VSMCs markedly increased the MMP 1 levels and activities of the culture media.MMP 1 levels were maximum when the ratio of both cells was 1.0.Separated coculture experiments revealed that the direct contact of both cells and cytokines contributed to MMP 1 production. Conclusion:Cell to cell interaction between monocytes and VSMCs enhanced MMP 1 synthesis.This implied that local MMP 1 production and activities induced by a monocyte VSMCs interaction may be play an important pathogenic role in the atherosclerotic plaque rupture.
