摘    要:本文通过毒力生物表型、抗原表达及相关基因结构的研究,对痢疾双价工程菌株FS-18、FS-15A及其回变株FS-15V安全保护的基础及遗传背景进行了分析。认为工程菌株同时表达福氏及宋内氏O抗原是其双价保护的基础。FS-18及其亲株48025A质粒缺失20Kb左右的侵袭相关基因片段,不表达四种侵袭相关蛋白,丧失侵袭生物表型是其安全与无毒力的分子基础。FS-15A与FS-15V侵袭基因完整,但Tn5插入位点的不同可能是它们毒力表型不同的原因。该研究为工程菌苗的改进提供了依据。

Abstract:The basis of safety and protection of Shigella bivalent hybrid-strains F-18.FS-15A and FS-15V was analysed by comparing their biological phenotypes,antigen expression and related gene fragments, and some useful information for constructing and improving Shigella genetic vaccine was got. Mixed LPS of S. flexneri and S. Sonnei was expressed on the surfaces of the three bivalent strains. and this accounted for their protection for S. flexneri and S. Sonnei. FS-18 deleted about 20kb invasive plasmid gene, had no Ipa phenotypes. This was its molecular basis of safety. FS-15A also undergone about 20kb deletion in large plasmid, no invasive gene deletion was found in FS-15A and Fs-15V. Ipa ABCD was expressed on the surfaces of FS-15A and FS-15V. FS-15A was avirulent, while FS-15V was virulent. The reason was discussed. The site and number of inserted Tn5 in the large plasmid of FS-15A was different from FS-15V.
