维甲酸对树突状细胞增殖及细胞骨架微丝的调节作用

摘    要:目的观察药理剂量的全反式维甲酸对人单核细胞来源的树突状细胞增殖及细胞骨架微丝的调节作用。方法 rhGM-CSF1000U/mL、IL-4500U/mL诱导人外周血单核细胞向树突状细胞分化,在细胞分化过程中加入0.1μmol/L全反式维甲酸,相差显微镜下观察细胞的形态变化及集落形成,活细胞计数和MTT检测细胞的增殖。成熟树突状细胞经全反式维甲酸处理24h后,用FITC标记的鬼笔环肽检测细胞骨架微丝的形态和分布。结果细胞因子诱导的外周血单核细胞来源的树突状细胞具有典型的树枝状结构。在树突状细胞的分化过程中,全反式维甲酸作用组的树突状细胞的产量明显低于对照组(P0.05)。已经分化成熟的树突状细胞经维甲酸处理后细胞骨架微丝出现重排和分布不均。结论药理剂量的全反式维甲酸抑制树突状细胞的增殖和分化,并可能通过细胞骨架调节树突状细胞的功能。

Abstract:Objective To study the effect of pharmacological doses of all trans retinoic acid on the proliferation and cytoskeleton of denderitic cells from human peripherial monocytes.Methods The human monocytes were isolated from peripherial blood,and then stimulated with 1 000 U/mL recombinant human granulocyte-macrophage colony stimulating factor(rhGM-CSF) and 500 U/mL recombinant human interleukin(rhIL)-4 for 7 days with or without 0.1 μmol/L all trans retionic acid.The differentiation process of dendritic cells was observed dynamically under the phase contrast microscope.Live cell counting and MTT assay were applied to evaluate the cell growth.The cytoskeleton microfilaments of mature dendritic cells treated with retinoic acid for 24 h were stained with FITC labelled phalloidin.Results The cytokins induced-peripherial blood monocytes derived-dendritic cells had typical dendritic morphology.Exposing cells during the process of differentiation to all trans retinoic acid resulted the reduced dendritic cell proliferation when compared with control group(P<0.05).The cytoskeleton microfilaments were rearranged in mature dendritic cells after exposed to all trans retinoic acid.Conclusion All trans retinoic acid with pharmacological concentration inhibites the proliferation and differention of dendritic cells from peripherial blood monocytes,and possibly take part in the regulation of the cytoskeletal structure of dendritic cells.
