摘    要:目的 探讨供者凋亡淋巴细胞对大鼠原位肝移植免疫排斥的影响.方法 用荧光标记供者(SD大鼠)脾淋巴细胞,诱导凋亡后,经尾静脉输入受者Lewis大鼠,观察荧光细胞在体内分布.取SD大鼠脾淋巴细胞诱导凋亡后输入Lewis大鼠,7 d后行大鼠SD-Lewis原位肝移植,观察术后第7天肝功能(ALT、TBiL) 的变化、病理改变...

Effect on immunological rejection by infusion of apoptotic spleen cells from in vivo donor rats after orthotopic liver transplantation
Abstract:【Abstract】 Objective To study Effect on immunological rejection of rat s receiving liver transplantation after infusion apoptotic cells from donors 1 week before the transplantation. Methods The collected apoptotic spleen cells were labeled with 5,6-carboxyfluorescein diacetate succinimidyl ester(CFSE)and the infused into Lewis rats via caudal vein, the distributions of cells in livers, spleen, lung were detected by fluorescence microscope in 30min、60min、and 12h after liver transplantation. at donor apoptotic cells were injected to recipients prior to SD-Lewis liver transplantation at day 7 . The blood levels of ALT and TBil were determined and liver pathological changes and survival time observed. Results After infusion, most apoptotic cells from donors congregated in liver after 60min and reduced a little after 12h, however, only few of them existed in spleed and lung. After SD-Lewis liver transplantation , Survival time markedly longer and Liver function of group C, which received apoptotic cells, was remarkably improved as compared with that of group A, which didn’t received apoptotic infusion at 7d after operation. Immunohistochemistry showed that the acute immunological rejection was dramatically alleviated in group C. Conclusions The liver deserves attention for study of the mechanism of allografts tolerance induced by pre-infusion of apoptotic donor lymphocytes. Pre-infusion of apoptotic donor cells can alleviate acute immunological rejection after orthotopic liver transplantation.
