摘    要:目的 观察Nbn 基因神经特异性敲除(Nbn-CNS-del)小鼠海马星形胶质细胞发育的形态学变化,探讨Nbn 基因在小鼠海马星形胶质细胞发育中的作用。方法 采用星形胶质细胞标记物GFAP,应用免疫组织化学技术和体视学方法对生后(P)7、14、21 d 的Nbn-CNS-del小鼠海马进行系统观察和定量分析,以非基因敲除(Nbn-CNS-ctr)的同窝仔鼠为对照组。结果 P7~21 d,与对照组相比,Nbn-CNS-del小鼠海马CA区及齿状回各层GFAP阳性细胞失去正常星形结构,面数密度减小(P<0.05)。结论 Nbn 基因神经特异性敲除小鼠海马星形胶质细胞发育障碍,Nbn 基因可能在海马神经胶质细胞发育中起重要作用。

Effect of Nbn gene on the development of Hippocampus astrocytes
Abstract:Objective To investigate the role of Nbn gene on the development of hippocampus astrocytes in Nbn gene neuro-specific deleted (Nbn-CNS-del) mice by studying their morphological changes. Methods GFAP monocloned antigen, immunohistochemistry and stereological methods were used to analyse the morphological changes of hippocampus astrocytes in Nbn-CNS-del and Nbn-CNS-ctr on postnatal day 7, 14 and 21. Results Comparing with Nbn-CNS-ctr, the GFAP positive cells of Nbn-CNS-del mice hippocampus lost their normal structures and the profile numberical densities of GFAP positive cells were decreased(P<0.05). Conclusion The development of hippocampus astrocytes in Nbn-CNS-del mice was disordered and Nbn gene might play an important role in the development of hippocampus astrocytes.
