芦丁治疗急性痛风性关节炎的实验研究

摘    要:目的通过尿酸钠(monosodium urate monohydrate,MSU)晶体诱导的炎症动物模型,探讨黄酮类化合物芦丁对急性痛风性关节炎(acute gouty arthritis,GA)的治疗作用。方法选用48只健康雄性SD大鼠,随机均分为6组:空白对照组(A组),模型组(B组),吲哚美辛组(C组),低剂量芦丁组(D组),中剂量芦丁组(E组)和高剂量芦丁组(F组)。A、B组给予蒸馏水,C组给予吲哚美辛,D、E、F组给予不同剂量芦丁灌胃,每天1次,连续给药5天后采用MSU晶体制备大鼠GA模型。造模后继续给药2天,用丝线测定造模前及造模后2、4、8、12、24和48h大鼠的踝关节周径;造模后48h大鼠以戊巴比妥钠麻醉,腹主静脉取血,取肝脏和踝关节滑膜,制备血清和组织匀浆,采用硫代巴比妥酸(thio-barbituric acid,TBA)比色法测定脂质过氧化程度丙二醛(malondialdehyde,MDA)和一氧化氮(nitric oxide,NO)以及抗氧化酶:超氧化物歧化酶(superoxide dismutase,SOD)、过氧化氢酶(catalase,CAT)、谷胱甘肽过氧化物酶(glutathioneperoxidase,GSH-PX)的水平。结果造模后48h,吲哚美辛和芦丁能够显著降低GA模型大鼠的踝关节肿胀度及MDA和NO水平(P<0.05或0.01),提高SOD、GSH-PX、CAT活性(P<0.05或0.01)。结论芦丁对MSU晶体诱导大鼠GA有显著的改善作用,其机制可能与抗氧化作用有关。

Experimental Study of Rutin in the Treatment of Acute Gouty Arthritis

Abstract:Objective To study the treatment effect of flavonoid Rutin on acute gouty arthritis (GA) in the animal model of monosodium urate monohydrate (MSU) crystal-induced inflammations. Methods Forty-eight healthy female SD rats were enrolled and equally randomized to six groups: blank control (group A), model (group B), Indometacin group (group C), low-Rutin group (group D) and middle-Rutin group (group E) and high-Rutin group (group F). Groups A and B were given distilled water, group C were given Indometacin, groups D, E and F were given different doses of Ru- tin. Rutin, indometacin or distilled water was given orally, once daily, for five consecutive days. GA models were made using sodium urate crystal and medicating for two days. The circumference of the ankle joints was measured by tie line be- fore modeling and 2, 4, 8, 12, 24 and 48 hours after modeling. At 48 hours after modeling, the rats were anaesthetized with sodium pentobarhital and then sacrificed. Their blood was collected to isolate serum. The liver and synovial tissue of the right hind limb ankle joints were immediately removed and homogenized, the levels of lipid peroxidation malondialde- hyde (MDA) and nitric oxide (NO) as well as anti-oxidant enzyme including superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH PX) were observed with thiobarbituric acid (TBA). Results At 48 hours after modeling, Rutin could significantly relieve edema in rats and decline the levels of MDA and NO (P(0. 05 or 0. 01), as well as enhancing the antioxidant enzymes activity such as SOD, GSH-PX and CAT in serum, liver and synovial tissue (P〈0. 05 or 0. 01). Conclusion Rutin has remarkable effect on improving symptoms of acute GA induced by MSU crystals in rats, which might be related to its antioxidative effect.
