Rab25 siRNA对人卵巢上皮性癌细胞生长和增殖的作用

目的：通过RNA干涉抑制Rab25在人卵巢上皮性癌的过表达,观察Rab25基因的siRNA对肿瘤细胞的生长和增殖作用.方法：构建针对Rab25基因的siRNA重组质粒,稳定转染A2780细胞,通过RT-PCR检测Rab25的表达;MTT法观察细胞增殖,绘制细胞生长曲线;克隆形成实验检测肿瘤细胞的集落形成能力.结果：成功构建Rab25基因的siRNA,稳定转染靶细胞后可显著降低Rab25的表达,细胞生长速度减慢、增殖能力下降.结论：Rab25基因的siRNA对肿瘤细胞的抑制作用,为卵巢癌基因治疗开辟新途径.

Study of cell growth and proliferation in ovarian cancer by Rab25 small interfering RNA

Objective: By RNA interference to down-regulate Rab25 gene and study the effect of siRNA of Rab25 in cell growth and proliferation. Methods: According to Rab25 mRNA sequence in the Genebank, the plasmids expressing siRNA against Rab25 were constructed and stably transected into A2780 cells,RT-PCR,MTT method and colony forming test were applied to measure cell growth and proliferation. Results: Rab25 siRNA expression vector was successfully constructed ,cells transected with the plasmids expressing siRNA targeting Rab25 gene effectively decreased the expression of Rab25 and decreased cell growth,proliferation. Conclusion: Rab25 siRNA expression vector has been successfully constructed,which will facilitate further studies of Rab25 function and its application in the treatment of ovarian cancer.
