兔胚成骨细胞移植修复异体兔骨缺损的实验研究

摘    要:目的 将胎兔成骨细胞与明胶海绵复合体移植于异体兔的骨缺损处 ,以促进骨折愈合 ,从而为临床上异体成骨细胞移植治疗骨缺损奠定理论基础。方法 利用组织块法将从胎兔颅骨中分离培养的成骨细胞经体外繁殖并冻存复苏后与明胶海绵复合形成复合体回植于异体兔的胫骨钻孔缺损处作为实验侧 ,对侧以明胶海绵吸附生理盐水作为对照侧 ,分别于细胞植入后 1、3、5、7、9、11、15周行大体观察和组织切片检查 ,并于 1、4、7、9周行X线检查 ,比较其修复骨缺损的能力和免疫排斥反应。结果  (1)植入异体成骨细胞明胶海绵复合体未见明显的免疫排斥反应 ;(2 )实验侧较对照侧骨愈合时间明显提前 (约 2周 )。结论 胚胎成骨细胞修复异体骨缺损能促进骨愈合 ,且在修复过程中未见明显的免疫排斥反应。

Abstract:Objective Theosteoblastofrabbitembryoandgelationspongecomplexwastransplantedtothesiteof bonedefeatofallogeneicrabbitinordertopromoteboneunionandlayafoundationforrepairofbonedefectandpromotion ofboneunioninclinicalpractice .Methods Thecranialboneoftherabbitembryowasculturedwithfissuremess method ,thentheacquiredosteoblastswhichhadpropagatedinvitroandundergonefrozenstorageandresuscitationand gelationspongeweremadeintoacomplex .Thecomplexwasthentransplantedintothetibiabonedefectofallogeneicrab bitastheexperimentside.Gelationspongeabsorbingphysiologicalsalinewasusedinthecontrolatoralsideasthecontrol side.Micropathologyandhistologyexaminationweredoneatthefirst,third ,fifth ,seventh ,ninth ,eleventhandfiftieth weeks,X rayexaminationwasdoneatthefirst,fourth ,seventhandninthweeksaftertransplantationrespectively .Healing statesandimmunologicalrejectionreactionwerecomparedbetweentwosides.Results (1)Thetransplantationofthe complexofosteoblastandgelationspongecausesnoobviousimmunologicalrejectionreaction . (2 )Thehealingtimeof bonedefectintheexperimentsideisobviouslyearlierthanthecontrolside (abouttwoweeks) .Conclusion Embryonic osteoblastcanrepairthebonedefectandpromoteboneunion ,andnoobviousimmunologicalrejectionreactioncanbe seen .
