海马内M1和M2受体与学习记忆关系的研究

摘    要:目的 研究海马内M1和M2受体亚型对学习记忆的调节。方法 SD大鼠分为3组,海马内分别微重注射M1、M2反义寡核苷酸(MASODN),以海马内注射生理盐水作为对照组,观察药物对大鼠被动逃避反应步入潜伏期的影响。结果 海马内注射M2ASODN大鼠的步入潜伏期显著缩短,较对照组减少75%,海马内注射M2ASODN大鼠的步入潜伏期显著延长。结论 海马内M1受体可正向调节学习记忆,而M2受体负向调节学习记忆。

Abstract:Objective To investigate the effects of M 1 and M 2 muscarinic receptors in the hippocampus on the latency of step-through inhibitory avoidance task. Methods Eighteen adult male SD rats were divided into three groups (n=6 each). Steel guide cannulas aimed at the hippocampus were implanted; bilateral infusion of M 1 or M 2 ASODN (antisense oligodeoxynucleotides) was given 16 hours prior to the training and testing of step-through inhibitory avoidance task. Results Intrahippocampal injection of M 1 ASODN impaired the retention of memory for inhibitory avoidance, while intrahippocampal injection of M 2 ASODN facilitated the fear-activated memory. Conclusion M 1 and M 2 muscarinic receptors modulate learning and memory, with M 1 receptors enhancing and M 2 receptors suppressing learning and memory.
