摘 要: | 目的:建立岩黄连中不同部位脱氢卡维丁的高效液相色谱定量测定方法。方法:采用ZorbaxODS C18(250mm×4.6mm,5μm)色谱柱,以乙腈-0.05mol/L磷酸二氢钾溶液(用10%GKOH调pH至5.0)(85:15)为流动相,流速1.0mL/min,检测波长为347nm,柱温为室温。结果:脱氢卡维丁在2.51~75.36μg/mL(r=0.99999,n=7)范围内线性关系良好,平均回收率为99.86%,RSD为0.58%。结论:该法简单、快速、重现性好,适用于岩黄连中脱氢卡维丁的定量分析。

Abstract: | Objective:To establish the HPLC method for determination of the contents of dehydrocavidine in different parts of Corydalis saxicola Bunting. Methods: Using Zorbax ODS C18 column(250 mm 4.6 mm, 5 μm), acetonitrile-solution of potassium phosphate(0.05 mol/L, adjust pH to 5.0 with 10 %KOH) (85 : 15) as mobile phase. Flow rate was 1.0 mL/min and detecting wavelength was 347 nm. Results:The calibration curve for dehydrocavidine was linear in the concentration range of 2.51- 75.36μg/mL ( r = 0. 999 99, n =7). The average recovery was 99. 860//00 with RSD=0. 58~. Conclusion: This method is simple, rapid and with better reproducibility for determination of content of dehydrocavidine in Corydalis saxieola Bunting. |
