糖尿病肾病患者血浆动态蛋白质组学研究

目的探讨糖尿病肾病（DN）患者血浆差异蛋白表达情况,为寻找DN敏感血浆蛋白标志物提供依据。方法 4例确诊的DN患者（DN组）和4例健康人群（对照组）作为研究对象,应用荧光差异双向凝胶电泳分析两组血浆蛋白质组差异表达,以上调或下调1.5倍为截断值筛选出差异表达的蛋白质斑点进行质谱分析。结果成功构建DN患者血浆蛋白差异表达双向凝胶电泳图谱,并分析鉴定出6种差异表达蛋白;通过质谱分析和蛋白质数据库（UniProtKB/Swiss-Prot）比对鉴定出6种蛋白质,分别为补体C3、C4、凝溶胶蛋白前体、纤维蛋白原γ、载脂蛋白E和细胞骨架蛋白16。结论荧光差异双向凝胶电泳可鉴定筛选出DN患者与正常人群差异表达的血浆蛋白质,并为进一步筛选DN血浆蛋白标志物提供依据。

Dynamic proteomics of plasma in patients with diabetic nephropathy

Objective To investigate the differential expression of plasma protein in patients with diabetic nephropathy（ DN） and to provide a basis for finding diabetic nephropathy sensitive plasma protein markers.Methods Four DN patients and 4 healthy people were included in this study.The plasma of the included patients were extracted and tested for differential expression by using 2-DDIGE.Higher or lower than 1.5 times was considered as the cutoff filter to screen the differentially expressed protein spots which would be analyzed by mass spectrometry.Results We successfully constructed the two-dimensional gel electrophoresis profiles of plasma protein differential expression of patients with DN,and 6 different expression plasma proteins were identified.By mass spectrometry and UniProtKB /Swiss-Prot,we identified 6 proteins,including complement C3,C4,gelsolin precursor,fibrinogen γ,apolipoprotein E and cytoskeletal protein 16.Conclusion The differential expression plasma protein between DN patients and healthy people can by identified by bidirectional fluorescence difference gel electrophoresis,which provides the basis for further screening DN plasma protein markers
