Sysmex XE2100血细胞分析仪与流式细胞检测抗体生成细胞的相关性

目的:通过对自身免疫性溶血性贫血(autoimmune hemolytic anemia,AIHA)患者抗体生成细胞(antibodysecreting cell,ASC)的检测,分析Sysmex XE2100分析仪与流式细胞仪的相关性及散点图的分布特征。方法:检测65例AIHA患者外周血白细胞并绘制SSC-SFL通道散点图;BD FACScalibur流式细胞仪检测ASC;显微镜分类计数200个白细胞中的ASC作为最终判定依据。结果:9个ASC/200WBC,流式细胞仪检测350个/μl,XE2100为级分布;1~6个ASC/200WBC,流式细胞仪检测100~300个/μl,XE2100为级分布;而在XE2100为级分布时,显微镜检查为阴性,流式细胞仪检测50个/μl左右。结论:检测ASC,流式细胞仪结果为最佳,而XE2100虽然不能准确定量,但其高速高可信度的分布图像是临床应用的重要补充。

Relationship of Sysmex XE2100 and BD FACScalbur Flow Cytometry to Determine Antibody Secreting Cell in Periphery Blood of Patient with Autoimmune Hemolytic Anemia(AIHA)

Objective：To analyse the pertinence relation of Sysmex XE2100 and BD FACScalbur flow cytometry to determine the antibody secreting cell in periphery blood of patient with autoimmune hemolytic anemia （AIHA）. Method ：To determine the ASC in periphery blood and draft SSC-SFL scatter graph with sysmex XE 2100,and analyse the ASC with BD FACScalicur flow cytometry and count the type I atypical lymphocyte with microscope. Results： 1-4 ASC/200WBC with microscope,1004300 ASC//＊I with flow cytometry,mid density distribution with XE2100;〉6 ASC per high power field with microscope examination, 〉 350 ASC/μI with Flow cytornetry,high density distribution with XE2100 analyzer ;While there was tow density distribution with XE2100,it was negative results with micros-cope examination. But it was 50 ASC/μt with flow cytometry. Conclusion：For the determination of Asc,FCM is the best method. Although XE2100 is not quantitative determination method,it＇s the important plus for the high rate and high trustwor- thiness.
