补肾平喘膏方对支气管哮喘大鼠肺组织糖皮质激素受体的影响

摘    要:目的:观察支气管哮喘大鼠肺组织中糖皮质激素受体(glucocorticoid receptor,GR)的变化情况以及补肾平喘膏方对其的影响,以探讨补肾平喘膏方防治支气管哮喘的作用机制。方法:60只SD大鼠随机分为正常对照组、模型组、地塞米松组、膏方预防组、膏方治疗组和膏方防治(预防加治疗)组,每组10只。除正常对照组外,各组大鼠以卵白蛋白致敏并诱发哮喘模型。采用地塞米松或膏方治疗后,HE染色法检测大鼠肺组织病理学改变,免疫组织化学法检测肺组织中GR的表达情况。结果:模型组肺组织GR表达低于正常对照组(P〈0.05),膏方预防组、治疗组、防治组肺组织GR的表达较模型组明显上调(P〈0.05),与正常对照组比较,差异无统计学意义(P〉0.05)。其中膏方预防组GR的表达显著上升(P〈0.01),膏方各组与地塞米松组相比亦明显上升(P〈0.01)。结论:补肾平喘膏方可能通过上调哮喘大鼠肺组织中GR的水平治疗哮喘。

Abstract:Objective:To observe the effects of medicinal extract for tonifying kidney to relieve asthma on glucocorticoid receptor (GR) expression in rats with asthma,and to explore its mechanism in treating asthma.Methods:Sixty SD rats were randomly divided into normal control group,untreated group,dexamethasone group,and medicinal extract-prevented,medicinal extract-treated,and medicinal extract-prevented and-treated groups,with ten rats in each group.Asthma was induced by intraperitoneal injection of ovalbumin (OVA) and forced inhalation of atomized OVA.Expression of GR in lung tissues was detected by immunohistochemical method.Pathological changes of the lung tissues were observed by HE straining.Results:Expression of GR was lower in the untreated group than in the normal control group (P0.05).Expressions of GR in medicinal extract groups were up-regulated as compared with those in the untreated group and dexamethasone group (P0.05,P0.01),and there were no significant differences as compared with the normal control group (P0.05).Conclusion:Medicinal extract for tonifying kidney to relieve asthma can increase the expression of GR in lung tissues of asthmatic rats,which may be one of its mechanisms in preventing and treating asthma.
