硒化壳聚糖对K562细胞的作用及其与NF-КB信号分子的关系

摘    要:目的:研究硒化壳聚糖对慢性粒细胞白血病细胞株K562增殖的影响,并且探讨这种影响与NF-КB信号分子的关系。方法:MTT法检测硒化壳聚糖对细胞增殖的影响,AO/EB荧光染色用来观察药物对细胞的凋亡作用。Westernblot的方法检测蛋白含量的变化。结果:硒化壳聚糖对K562细胞抑制作用呈量效、时效关系,硒化壳聚糖200mg/L作用48h对K562细胞的抑制率高达90.7±10%;并使细胞出现明显的凋亡现象。Westernblot的实验结果表明硒化壳聚糖使K562细胞NF-КB蛋白含量明显减少,且呈量效关系。结论:硒化壳聚糖可减少进入细胞核内NF-КB的蛋白含量从而下调其下游基因表达,最终抑制K562细胞增殖,诱导细胞发生凋亡。

Effect of selenium chitosan on proliferation of K562 cells and down regulation of NF-КB

Abstract:Objective: To study the effect of selenium chitosan on proliferation of K562 cells and the relationship of the NF-КB. Methods: MTT was used to determine the proliferative effects of drug on tumor cells. AO/EB was used to determine the apoptosis of the drug treated cells. Western blotting was used to examine the abundance of signal protein molecules expressed by tumor cells. Results: Exposure of K562 cells to selenium chitosan produced both dose and time dependent increase in proliferative inhibition rate .The inhibition rate of K562 cells treated by selenium chitosan 200mg/L for 48h was as high as 90.7±10%. Selenium chitosan could induce tumor cell apoptosis . The abundance of NF-КB was down regulated in selenium chitosan treated K562 cells. Conclusions: Selenium chitosan can induce apoptosis and inhibit the proliferation of K562 cells and this correlates with down regulation of the abundance of NF-КB.
