摘    要:目的 采用原位杂交法观察反式脂肪酸和饱和脂肪酸诱导糖尿病大鼠肝脏组织趋化素(chemerin)的表达情况及相关因素的影响,探讨炎症、脂代谢在肥胖、胰岛素抵抗及2型糖尿病发生发展中的作用.方法 将32只健康雄性SD大鼠随机分成4组:正常对照组(NOR组)8只,生理盐水灌胃;饱和脂肪酸组(SFA组)8只,脂肪乳灌胃;反式脂肪酸组(TFA组)8只,人造奶油灌胃;糖尿病组(DM组),8只.观察空腹血糖(FPG)、游离脂肪酸(FFA)、超敏C反应蛋白(hs-CRP)和血脂等指标.原位杂交法检测大鼠肝脏组织chemerin的表达情况,并比较chemerin与其他相关指标的关系.结果 SFA、TFA组大鼠体重较喂养初期增加,DM组大鼠体重较NOR组降低明显(P<0.05).血清FINS水平SFA、TFA、DM组均高于NOR组(P<0.01).hs-CRP、FFA的测定SFA、TFA、DM组均高于NOR组(P<0.05).大鼠肝组织chemerin mRNA的表达SFA、TFA、DM组高于NOR组(均P<0.01).chemerin分别与HOME-IR(r=0.544,P<0.05)、ISI(r=-0.544,P<0.05)、hs-CRP(r=0.575,P<0.05)、TG(r=0.7,P<0.05)、LDL-C(r=0.77,P<0.01)具有相关性.结论 Chemerin在早期能在一定程度上反映胰岛素抵抗的程度,加重脂肪细胞的功能障碍并对肝脏组织造成影响,为一致炎因素.

Abstract:Objective Using in situ hybridization method to observe the expression of chemerin and influence of related factors,which from different diets induced insulin resistance and IL-6 intervention diabetic rat liver tissue.To explore the effect of inflammation and lipid metabolism in obesity,insulin resistance and the development of incident diabetes.Methods 32 healthy SD male rats were randomly divided into 4 groups,8 rats in each group.Normal control group(NOR group)8,normal saline;saturated fatty acid group(SFA group)8,intralipid gavage;trans-fatty acid group(TFA group)8,margarine gavage;diabetes mellitus control group(group DM)8.Until the 8th week,the animals were killed,fasting plasma glucose(FPG),fasting insulin(FINS),free fatty acids(FFA),high sensitive CRP(hs-CRP)and lipids were measured.The expression of chemerin on the liver of the rats were tested by in situ hybridization.Results Compared with the initiation,the rats'body weight of SFA、TFA groups increased than NOR group(P0.05).Serum FINS levels of SFA,TFA,DM groups were higher than the NOR group(P0.01).Hs-CRP,FFA in the SFA,TFA,DM groups were higher than the NOR group(P0.05),and chemerin was positively correlated with HOME-IR(r=0.544,P0.05),LDL-C(r=0.77,P0.01),hs-CRP(r=0.575,P0.05)and TG(r=0.7,P0.05),negatively correlated with ISI(r=-0.544,P0.05).Conclusions We found that chemerin expression level was high in insulin resistance period,though its expression level decreased in type 2 diabetes periods,but was still higher than normal level.Chemerin can somewhat reflect the insulin resistance level early.It can also aggravate the dysfunction of fat cells and affect the liver tissue,but the mechanism of its interaction with other factors needs further research.
