过敏性紫癜患者外周血细胞间黏附分子2、3与黑色素瘤细胞黏附分子表达的初探

摘    要:目的 检测细胞间黏附分子2(ICAM-2)、ICAM-3、黑色素瘤细胞黏附分子(MCAM)在过敏性紫癜(HSP)患者外周血的表达,并探讨其在本病发病机制中的意义.方法 采用双抗体夹心法(ELISA)检测60例HSP患者组与50例正常对照组血清中ICAM-2、ICAM-3水平.采用实时荧光定量RT-PCR法检测HSP患者及对照组外周血单个核细胞(PBMC)中MCAM mRNA的相对表达.结果 HSP患者血清ICAM-2、ICAM-3水平明显高于正常对照组,差异均具有统计学意义(P<0.05),患者各组间两两比较,血清中ICAM-2、ICAM-3水平差异无统计学意义(P≥0.05).患者组PBMC中MCAM mRNA的相对表达量明显高于正常对照组,差异具有统计学意义(P<0.05),患者各组间两两比较,差异无统计学意义(P>0.05).结论 ICAM-2、ICAM-3、MCAM可能在HSP的发病中起重要作用.

Abstract:Objective To investigate the expression and significance of ICAM-2,ICAM-3 and MCAM in peripheral blood of patients with anaphylactoid purpura(HSP).Methods Sixty patients with HSP and fifty normal human controls were included in this study.ELISA was performed to measure the level of serum ICAM-2,ICAM-3.SYBR Green Ⅰ real-time RT-PCR was performed to detect the mRNA of MCAM in PBMCs.Results The levels of serum ICAM-2,ICAM-3 were significant higher in patients with HSP than in normal human controls(P0.05).No significant difference was observed in the levels of ICAM-2 and ICAM-3 among patients with different types of HSP(all P0.05).The levels of MCAM mRNA in PBMCs were significantly higher in HSP patients than in normal human controls(P0.05),no significant difference was observed in the levels of MCAM mRNA among patients with different types of HSP(all P0.05).Conclusions ICAM-2,ICAM-3 and MCAM may be associated with the pathogenesis of HSP.
