摘    要:目的 比较两种不同脓毒症小鼠模型,探讨两种模型的差异,以及各自在脓毒症研究中的意义.方法 取60只雄性BALB/c小鼠,随机分为假手术组、盲肠结扎穿刺(CLP)组和急性腹膜炎(AP)组,观察不同时间点各组的存活率,流式细胞术(FCM)检测脾脏Gr-1+/CD11b+细胞比例,ELISA检测外周血细胞因子水平.结果 AP组生存率在术后早期(术后1~2 d)明显低于CLP组,但后期(术后2 d以后)二者无统计学差异.CLP组脾脏Gr-1+/CD11b+细胞比例在术后第7天和第21天均高于AP组.外周血细胞因子(IL-6、IL-10)表达水平,AP组在术后6 h和12 h明显高于CLP组,但在术后24 h二者无统计学差异.结论 两种模型均较好地模拟了脓毒症的病理机制,但CLP更适合研究腹腔脓肿相关疾病引起的脓毒症,而AP模型则更类似于急性弥散性腹膜炎相关疾病所致的脓毒症.

Abstract:Objective To investigate the difference between two distinct animal models for polymicrobial sepsis in mice as well as their difference in mimicking sepsis. Methods Sixty male BALB/c mice were randomly divided into three groups which were SHAM group, CLP group and AP group. The survival rate of several time points 'after surgery was observed in each group. The percentage of Gr-I +/CD11 b+ cells was determined by flow cytometry (FCM) ,and the serum eytokines(IL-6 and IL-10)was detected by ELISA. Results The survival rate of AP group was significantly lower than the survial rate of CLP in the early period( 1 d or 2 d after surgery) ,but the survial rate showed no difference between AP group and CLP group in the late period( beyond 2 d after surgery). The percentage of Gr-1 +/CDllb+ ceils in spleen of CLP group was higher than that of AP group on both the seventh day and the twentieth day after surgery. The serum IL-6 and serum IL-10 of AP group was significantly higher than that of CLP group at the 6 h and the 12 h after surgery, but no difference was showed at the 24 h after surgery between the AP group and the CLP group. Condusions Both the two animal models mimic the process of sepsis properly. However, the process of CLP model resembles peritoneal abscess while the process of AP model is similar to acute diffuse peritonitis.
