摘    要:目的 探讨12-脂加氧酶(12-LO)在病毒性心肌炎(VMC)小鼠心肌内的表达.方法 将4周龄Balb/c小鼠随机分成实验组和正常对照组:采用柯萨奇病毒亲心株B3(CVB3)感染Balb/c小鼠建立VMC小鼠模型;通过RT-PCR法测定外周血12-脂加氧酶mRNA(12-LO mRNA),比色法测NO的变化,化学发光免 疫法检测血清心肌肌钙蛋白I(cTnI),HE染色观察心肌损害程度.结果 (1)实验组12-LOmRNA表达于CVB3感染第3天(0.623±O.321)、第7天(1.311±0.208)、第14天(1.018±0.302)升高,与对照组(O.401 ±0.014)比较差异有统计学意义(P<0.01),第28天(0.512±0.025)虽逐渐下降,但与对照组(0.401 ±0.014)比较差异仍有统计学意义(P

Abstract:Objective To examine the myocardial expression of 12-1ipoxygenase (12-LO) in mice with viralmyocarditis. Methods One hundred 4 weeks Balb/c mice were randomly divided into two group:normal control group and experiment group .Viral myocarditis model was established by subjecting Balb/c mice to Coxsackie virusB3( CVB3 ) infection. The heart tissues were examined at 3,7,14,28 days after CVB3 infection.Expression of 12-LOmRNA was determined by RT-PCR. NOwas measured by colorimetry methods, eTnI was measured by chemicalluminescence immunoassay. Myocardial damage was assessed by HE staining. Results ( 1 ) The expressions of 12-LOmRNA in experiment group were 0. 623±0. 321,1. 311±0. 208,1. 018±0. 302 ,and 0. 512±0. 025 ,respectively ,at 3,7,14,28 days. The difference between two groups was significant at 3,7,14,28 days. (2) The expressions of NOin experiment group were (42. 38±8. 84 )μmol/L, (70. 25±11.62 )μmol/L, ( 68. 11±9.78 ) I.t.mol/L, and (45.35±11.12 )μmol/L respectively at 3,7,14,28 days,the difference between two groups at each time point was significant (P〈0.01). (3)The expressions of eTnI in experiment group were (7. 88±1.23 )μg/L, (1.98±0. 39) μg/L, (0. 98±0. 22 )μg/L at 3,7, 14 days, the difference between two groups at each time point was significant (P 〈0. 01 ). (4) The expression of 12-LO mRNA in the experimental group was significantly correlated with cardiac pathology score (r =0. 317 ,P 〈 0. 05 ). (5) NO expression in the experimental group was significantly correlated with cardiac: pathology score ( r = 0. 436, P 〈 0. 05 ). ( 6 ) The expression of 12-LO mRNA and NO in theexperimental group were significantly correlated with the expressions of cTnI ( r = 0. 429 ;0. 421, P 〈 0. 05 ) ;.(7 ) The expression of 12-LO mRNA in experiment group was significantly correlated with NO expression ( r = 0. 531, P 〈 0. 05 ). Conclusions The expression of NO and 12-LO are found in mice with viral myocarditis, and they are positively associated.
