人卵巢粘液性囊腺癌细胞系的建立及生物学特性的研究

摘    要:目的 探讨人卵巢粘液性囊腺癌细胞系的建立,并研究其生物学特性。方法 用贴壁培养法建立细胞系,冷冻保存。用光学显微镜、电子显微镜观察细胞系的形态,研究细胞系的生长及生物学持性。结果 国内第1株人卵巢粘液性囊腺癌细胞系建立成功,命名为OMC685,现已传至91代,冷冻8年多复苏后仍生长良好。其生物学特性为,形态学观察证实该细胞系具有腺上皮癌细胞的特点;细胞生长旺盛;植物血凝素凝集试验阳性;染色体为亚三倍体,有畸变;异种动物移植阳性;免疫组织化学酸性粘液染色阳性;雌二醇及黄体酮阳性;无支原体及病毒污染;对脊髓灰质炎病毒I型、腺病毒7型及麻疹病毒敏感。结论 OMC685是一株独特的人卵巢癌细胞系。

Abstract:OBJECTIVE: To establish a cell line of human ovarian cancer, and study its characterization. METHODS: The cell line was established by the cultivation of subsides walls, and kept by freezing. The morphology was observed by microscope and electromicroscope. The authors studied its growth and propagation, the agglutination test of phytohemagglutinin (PHA), the chromosome analysis, heterotransplanting, immuno-histochemistry staining, the analysis of hormone, the pollution examination and the test of sensitivity to virus etc. RESULTS: A new human ovarian carcinoma cell line, designated ovarian mucinous cystadenocarcinoma 685 (OMC685), was established from mucinous cystadenocarcinoma. This cell line had subcultured to 91 generations, and some had been frozen for 8 years and revived, still grew well. This cell line possessed the feature of glandular epithelium cancer cell. The cells grew exuberantly, and the agglutinating test of PHA was positive. Karyotype was subtriploid with distortion. Heterotransplantations, alcian blue periobic acid-schiff (AbPAS), mucicarmine, alcian blue stainings, estradiol (E2) and progesterone were all positive. Without being polluted, it was sensitive to polivirus-I, adenovirus 7 and measles virus. CONCLUSIONS: OMC685 is a distinct human ovarian tumous cell line.
