骨髓干细胞肝移植对肝硬化模型猪并发心电图改变的影响

摘    要:目的探讨骨髓干细胞(BMSCs)肝移植对肝硬化模型猪并发心电生理和心电图改变的影响。方法选择健康湖北白猪(3~4月龄)60头,将其随机分成两组,对照组(12头)普食喂养;实验组采用四氯化碳诱导建立猪肝硬化动物模型(48头)。自体BMSCs采集、分离与移植;在BMSCs肝移植前后对肝硬化模型猪进行心电图检测。结果①模型猪肝脏超声体积缩小、呈结节样改变,肉眼观食管胃底静脉明显曲张,镜下观假小叶形成,判定猪肝硬化动物模型制作成功;②48例肝硬化模型猪BMSCs肝移植前出现心电图异常27例(56.25%),其中异常心电图以长QTc间期发生率37.03%(10例)为最高;③BMSCs肝移植后12周,心电图异常14例(占29.17%),长QTc间期发生率4例次(占28.57%)。结论肝硬化模型猪心电图异常率明显升高,主要表现为QTc间期延长,且随肝损害严重程度加重而延长;BMSCs肝移植后,心电图异常率以及长QTc间期发生率明显下降,心电生理得到改善;BMSCs肝移植本身可诱发肝硬化模型猪出现暂短心电生理紊乱。

The influence of intrahepatic transplantation of bone marrow stem cells on electrocardiogram in the porcine model of liver cirrhosis

Abstract:Objective To investigate the influence of intrahepatic transplantation of bone marrow stem cells(BMSCs) on electrocardiogram(ECG) in the porcine model of liver cirrhosis.Methods Sixty Hubei white pigs(3~4 months) were randomly divided into experimental group(n=48) and control group(n=12).Carbon tetrachloride was used to induce liver cirrhosis in experimental group.Results ① The porcine liver cirrhosis model was established,which was indicated by small and nodular liver in ultrasonic examination,and esophagogastric varices and pseudo-lobule formation.②ECG abnormalities were found in 27 of 48 model pigs(56.25%) before transplantation,and the incidence rate of long QTc interval was highest(37%,10 cases).③ECG abnormalities were found in 14 cases(29.17%),and QTc interval prolonged in 4 cases(28.57%) 12 weeks after transplantation.Conclusion ECG abnormal rate increased significantly in porcine model of liver cirrhosis and the main abnormality is extended QTc interphase which is associated with the severity of liver damage.ECG abnormal rate and long QTc interphase rate declined obviously after intrahepatic BMSCs transplantation.Intrahepatic BMSCs transplantation itself may induce temporary ECG disorders in the porcine model of liver cirrhosis.
