白果内酯对半乳糖诱导的痴呆小鼠学习记忆功能的保护作用

摘    要:目的研究白果内酯(Bilobalide,BB)对D-半乳糖诱导的痴呆小鼠的保护作用。方法采用D-半乳糖皮下注射建立小鼠衰老模型,通过跳台和避暗实验检测小鼠的学习和记忆能力;采用比色法,测定小鼠脑组织中超氧化物歧化酶(SOD)活力和丙二醛(MDA)含量;采用HE染色法,观察小鼠脑组织病理形态学变化。结果结果表明,白果内酯(5、10mg/kg)能不同程度地提高痴呆小鼠的学习记忆成绩,提高脑组织中SOD活性,降低MDA含量,HE染色结果提示白果内酯对小鼠脑组织皮层和海马CAl区病理形态学变化有一定的改善作用,结论白果内酯对半乳糖诱导的痴呆小鼠有一定的保护作用,其作用机制可能与抗氧自由基有关。

Protective effects of Bilobalide on learning and memory in dementia mice induced by D-galactose

Abstract:Objective To investigate the protected effects of Bilobalide(BB) on dementia mice induced by D-galactose.Methods The mode of dementia mice established by subcutaneous injecting D-galactose,BB(2.5,5,10 mg·kg-1) were taken by intragastric administration.Step-through test and step-down test were used to evaluate the effects of BB on learning and memory in mice.The superoxide dismutase(SOD) activity and malondialdehyde(MDA) level of brain tissue were detected.Morphology of hippocampal CA1 area and cerebral cortex was observed through HE staining.Results BB(2.5,5,10 mg·kg-1) differently enhanced the ability of learning and memory in dementia mice,significantly increased SOD activity and decreased MDA content.The results of HE staining showed that BB could improve the pathological changes in hippocampal CA1 area and cerebral cortex.Conclusion BB can protect against dementia mice induced by D-galactose,and the mechanism may be related with antioxidant.
