A-NK细胞抗肝癌Walker256瘤株的实验研究

目的:观察A-NK细胞的体外生长与增殖,以及杀伤肿瘤细胞的能力,研究A-NK细胞局部注射的体内抗肿瘤作用。方法:用淋巴细胞分离液分离单个核细胞(PBMC),培养LAK细胞和A-NK细胞,分别将LAK细胞、A-NK细胞和Walker-256瘤株细胞接种于培养板中,培养24h后,用四甲基偶氮唑盐(MTT)方法测定吸光度,计算肿瘤杀伤率。复制鼠肝癌模型,分别将LAK、A-NK细胞经肝动脉注入鼠肝癌模型中,观察两组动物的生存时间。结果:A-NK细胞的增殖明显快于LAK细胞(P<0.05),A-NK细胞对肿瘤细胞的杀伤活性比LAK细胞强(P<0.01),A-NK细胞能显著延长肝癌动物模型的生存期,与LAK细胞比较,具有显著性差异(P<0.01)。结论:A-NK细胞具有增殖快,抗瘤活性强,在体内能抑制肿瘤生长,延长荷瘤动物的生存期。

The experimental research on local application of A-NK cells in anti-tumor

Objective:By observing the external growth and hyperplasia of A-NK cells and the ability of killing tumor cells,to study the functions of internal anti-tumor by local injection of A-NK cells.Methods:To use Lymphocytes separation medium to separate the single nuclear cell(PBMC),to develope LAK cells,A-NK cells and then inoculate respectively the LAK cells,the A-NK cells and Walker-256 cells on culture boards for 24 hours,use the measurement of MTT to determine the absorbance and figure out its kill probability of tumor.Copy the models of rat liver cancer,respectively inject LAK,A-NK into them through arteria hepatica to observe the survival time of the two groups of the animals.Results:The hyperplasia of A-NK cells were obviously faster than that of LAK cells (P<0.05`),the kill activity of A-NK cells were stronger than that of LAK cells(P<0.01),and A-NK cells can obviously prolong the life span of the model of the liver cancer animals,compare with the LAK cells,it had significant difference(P<0.01).Conclusion:A-NK cells have the characteristic of hyperplasia quickly,anti-tumor activity strongly,it can restrain the growth of the tumor in the body and prolong the existence period of the tumor-bearing animals.
