摘    要:目的 探讨雌性大鼠老化过程中血清雌激素浓度、脾细胞雌激素受体( E R) 含量及白细胞介素2( I L2) 、肿瘤坏死因子( T N F) 活性变化的相关性。方法 以各月龄组雌性大鼠为研究对象,应用放射免疫法、放射配体结合分析法、 M T T 法及细胞毒法等技术,观察上述各指标随老化的变化。结果 随着老化雌性大鼠血清 E2 浓度与脾细胞 E R 含量及脾细胞诱生 I L2 、 T N F 活性均明显下降,并呈明显正相关。结论 生理水平血 E2 对脾细胞 E R 有正向调节作用,雌激素可能通过免疫细胞 E R 来调节免疫功能。随着老化到来,雌激素水平下降及免疫细胞 E R 含量降低,这可能是老年机体免疫功能衰退的原因之一。

Abstract:Objective To investigate the relationship between serum E 2, splenic estrogen receptor (ER) and IL 2, TNF levels in aging female rats. Methods In aging female rats, ER proteins of splenic cells were determined with radio ligand binding assay, estrodial concentration of serum with RIA, and activities of IL 2 and TNF with MTT and cytotoxic methods. Results With aging, levels of serum E 2 and ER of splenic cells and IL 2 and TNF activities decreased progressively in female rats. Positive correlation existed between levels of serum E 2 and splenic ER and between levels of splenic ER and IL 2 and TNF activities in aging female rats. Conclusions Splenic ER is generally up regulated by serum estrogen level. Estrogen may regulate the function of immune system via ER on immune cells. In aging rats, decrease in serum E 2 leads to fading of ER in immune cells and may results in low immunity.
