联合低温保护剂对脐血造血干细胞长期低温保存的作用

摘    要:目的 观察联合低温保护剂对脐血造血干细胞长期低温保存的作用。方法 60份由联合低温保护剂(5%DMSO+6%HES+5%人体白蛋白)保存在岭南脐血库的脐血,按时间分为1,2,3,4年等4组,复温后观察其细胞活力、总有核细胞数(TNC)、CD34^ ,CD34^ CD38^-,CFU-GMey BFU-E的回收率。结果 60例脐血冻存后的细胞活力为96.39%,TNC,CD34^ ,CD34^ CD38^-,CFU-GM及BFU-E的回收率分别是95.89%,94.45%,88.88%,87.77%及83.18%,不同时间组的上述指标之间差异无显著性(P>0.05)。结论 联合低温保护剂具有效果可靠、操作简单等优点,适用于脐血造血干细胞的长期低温保存。

Abstract:Objective To observe the protection of combined cryoprotectant (5%DMSO+6%HES+5%human albumin) on umbilical blood stem cells stored for different cryopreservation duration. Methods 60 cryopreserved umbilical bloods were divided into 4 groups according to duration(1, 2, 3 and 4 years). The viability of cells, total nucleate cells (TNC),CD 34 +, CD 34 +CD 38 -,CFU-GM, and BFU-E were tested before and after cryopreservation with combined cryoprotectant (5%DMSO+6%HES+5%human albumin) . Results The viability of cells,TNC,CD 34 +, CD 34 +CD 38 -,CFU-GM, and BFU-E in 60 cryopreserved umbilical bloods were 96.39%, 95.89%, 94 45%, 88 88%, 87 77% and 83 18% No significant difference were found among different cryopreservation duration groups( P >0 05). Conclusion With the advantages of reliability and simplicity, combined cryoprotectant is desirable long term cryopreservation of umbilical cord blood stem cells
