摘    要:目的探讨胸苷激酶(TK)基因在垂体腺瘤治疗中的作用及其机制。方法应用免疫印迹法,检测AdGHTK质粒转染GH3细胞后TK基因的表达情况;使用非放射性细胞增殖检测法,测定不同感染复数(MOI)AdGHTK转染后的GH3细胞对更昔洛韦的敏感性;将转染AdGHTK的GH3细胞和未转染细胞以不同比例混合,观察旁观者效应。结果TK基因可转染入GH3细胞并成功表达;即使在低MOI下,转染组细胞成活率也随更昔洛韦浓度的增加而降低;存在旁观者效应。结论将TK基因导入GH3细胞并经更昔洛韦处理,可明显降低肿瘤细胞存活率,为垂体腺瘤的治疗提供了可靠的理论依据。

Abstract:Objective To explore the efficacy of thymidine kinase (TK) gene for pituitary adenoma. Methods Applying mmunoblotting, the expression of TK gene was detected after AdGHTK plasmid transfect GH3 cells; and the sensitivity of CH3 cells was determined by non-radioactive cell Proliferation assay, which had been transfected by AdGHTK with different multiplicities of infections (MOls) to the ganciclovir; GH3 cells transfected with AdGHTK were mixed with the non-transfected cells in different ratios to observe the bystander effect. Results TK gene could be transfected into GH3 cells and expressed successfully; even in the condition of lower MOl, the survival rate of cells in transfection group kept decreasing as the concentration of ganciclovir increased. In addition, there existed a bystander effect. Conclusion Transfection of TK gene into GH3 cells and the treatment with GCV obviously reduced the survival rate of tumor cells, which provides a reliable theory basis for the treatment of pituitary adenoma.
