吡格列酮对大鼠脑出血后Cyt-C表达影响及意义

摘    要:目的 探讨吡格列酮(Pio)对大鼠脑出血后细胞色素C(Cyt-C)表达的影响及其意义。方法 将SD大鼠随机分为假手术组、对照组、Pio干预组(Pio组)、Pio与5羟基葵酸盐(5-HD)干预组(5-HD+Pio组),每组8只。应用自体血尾状核定位注射方法建立大鼠自体血脑出血模型。采用RT-PCR和Western Blot方法检测各组大鼠血肿周围组织Cyt-C mRNA和蛋白的表达情况。结果 与假手术组比较,对照组、Pio组、5-HD+Pio组Cyt-C表达明显升高(F=55.053、854.023,t=10.681-43.556,P〈0.05);Pio组Cyt-C表达明显低于对照组、5-HD+Pio组(t=4.794-25.357,P〈0.05);对照组与5-HD+Pio组Cyt-C表达水平差异无显著性(P〉0.05)。结论 Pio可以降低大鼠脑出血后Cyt-C的表达,从而减少神经细胞凋亡,其神经保护作用可被5-HD所拮抗。

Abstract:Objective To explore the effects of pioglitazone on expression of Cyt-C after intracerebral hemorrhage (IH) in rats. Methods Sprague-Dawley rats were randomly divided into the following four groups: sham-operated group, control group, Pioglitazone (Pio) treatment group, 5-HD+Pio treatment group, with eight rats in each group. Application of autoblood caudate nucleus injection, a model of cerebral hemorrhage in rats was created. The expression of Cyt-C in tissues around the hematoma was detected by RT-PCR and Western Blot. Results Compared with the sham-operated group, the expression of Cyt-C in the control group, Pio group, and 5-HD+Pio group was obviously higher (F=55.053,854.023;t=10.681-43.556;P〈0.05); the expression of Cyt-C in Pio group was lower than that in the control and 5-HD+Pio groups (t=4.794-25.357,P〈0.05); the difference of Cyt-C expression between the control and 5-HD+Pio groups was not significantly different (P〉0.05). Conclusion Pioglitazone can reduce the expression of Cyt-C in rats after intracerebral hemorrhage, and thereby decrease apoptosis of nerve cells, its neuroprotective effect can be antagonized by 5-HD.
